The impact of exosomes derived from bone marrow mesenchymal stem cells on CDK4 gene expression levels, and the proliferation and migration ability of breast cancer cells

Abstract
Breast cancer (BC) is the most common cancer and the leading cause of death from cancers among women. Cyclin-dependent kinase 4 (CDK4) is one of the most important genes regulating the cell cycle and acts as an oncogene, so that its expression level increases in various types of cancer, including breast cancer. Increasing evidence suggests that exosomes secreted by mesenchymal stem cells (MSCs) inhibit tumor growth by transducing signaling molecules. Therefore, the aim of this study was to determine the expression of CDK4 gene and the rate of cell proliferation and migration in breast cancer cell line 4T1 under the influence of treatment with exosomes secreted by bone marrow mesenchymal stem cells. 4T1 cells were cultured and treated with 100 μg/ml of exosomes. The change of CDK4 gene expression levels was evaluated by real-time PCR. Also, the rate of proliferation and migration rate of cells were measured. Our results showed that treatment of 4T1 breast cancer cells with exosomes derived from bone marrow mesenchymal stem cells decreased the proliferation rate and inhibited cell migration compared to the control group. Also, it led to a significant decrease in CDK4 gene expression as a key regulator of the cell cycle. In conclusion, Exosomes derived from mesenchymal stem cells can significantly reduce the expression of the CDK4 gene and ultimately lead to the reduction of cancer cell proliferation and migration.
