Abstract
Barley is a diploid plant with 14 chromosomes, and the evaluation of its genotypes' kinship provides useful information to breeders. In the present study, the genetic diversity of 18 barley genotypes was evaluated using the ISSR molecular marker. After extracting leaf DNA and ensuring the desired quantity and quality, a polymerase chain reaction was performed using ISSR primers. In this experiment, the 12 primers used produced 180 bands, 159 of which were polymorphic. The percentage of polymorphism (PIC) for all primers was 88.37% and the average number of polymorphic bands was 15.9 bands. The highest and lowest PIC values were related to primers F1 and F12, respectively. Primer F1 showed the highest and primer F12 the lowest amount of heterozygosity. The marker index (MI) was the highest in primer F1 and the lowest in primer F5. The results of this research confirmed the high efficiency of the ISSR marker in examining the diversity and molecular grouping of barley cultivars and showed that there is a suitable genetic diversity between the studied barley genotypes and they can be used in breeding programs.
