چکیده
با توجه به اهمیت انکوزومهای سرطانی در پیشروی سلولهای سرطانی، دو مدل غشای انکوزوم ایستا(صلب) و پویا(سیال) ساخته شد و نقش درجهی اشباع دو لایهی لیپیدی، بواسطه ایجاد تنوع در دنبالههای اسیدچرب واجد چند پیوند دوگانه در مدل پویا، با پپتید GL13K و توسط شبیه سازی دینامیک مولکولی بررسی شد. تراجکتوریهای شبیهسازی، ناهمواریهای سطحی بیشتر انکوزوم پویا را نسبت به ایستا نشان داد. فسفولیپیدها در مدل ایستا بسته بندی بهتری نسبت به پویا دارد. مقایسهی ماژولهای تراکم پذیری سطحی در هر دو غشاء مقاومت بیشتر مدل ایستا را در برابر نیروهای خارجی نسبت به مدل پویا، نشان داد. نتایج حاصل از آرایش اسیدهای چرب اولئیک بعنوان فراوانترین نوع اسیدچرب در هر دو غشای مدل، سیالیت مدل پویا نسبت به ایستا را تأیید کرد. انکوزوم پویا با فراهم کردن فضای سطحی بیشت برای جذب سطحی و نفوذ پپتید، تعاملات پایدارتری را شکل داد. تعاملات وابسته به ساختار مارپیچ آلفا در مدل ایستا تا 50 نانوثانیه ابتدایی بوده ولی درمدل پویا تا 175 نانوثانیه حفظ شد و در ادامه پپتید ساختار خود را از دست داد. رصد برهم کنشها در طی بازههای زمانی 25 نانوثانیهای، نقش کلیدی رزیجوهای KIIKLKA و آمینو اسید لیزین 5 را نشان داد. فسفولیپیدهای SDPE و SAPE در مدل پویا نقش ویژهای در ثبات برهم کنشها دارد. مقایسهی نیروهای دینامیک مولکولی هدایت شده تمایل بالای جذب سطحی پپتید به دو لایهی مدل پویا را بواسطهی آنالیزهای هدایت شده تأیید کرد. پژوهش حاضر با نتایج آزمایشگاهی مبنی بر خواص ضد سرطانی پپتید GL13K هم راستا می باشد.

Molecular dynamic simulation provides computational insights into membrane alterations during interaction with peptides and chemical drugs. In such a case, modeling realistic bilayers is critical because the peptide-membrane interactions depend on bilayer lipid compositions. According to these facts, in this project two cancerous bilayers as static(solid) and dynamic(fluid) models, constructed and the effect of saturation degree on bilayer function was analyzed by incorporating polyunsaturated fatty acids on bilayer hydrophobic core. Then their interaction with GL13K peptide were evaluated using All-atom and Steered molecular dynamic simulations. Based on results, dynamic model, provides acceptable surface area for primary interaction, surface adsorption, and peptide penetration during 200ns than static one. The persistence of these interactions in static models changed after 50 ns of simulation and after that time, the peptide lost its dominant helical structure. Steered MD simulation, also demonstrated the high binding affinity of peptide to the surface of dynamic model. Tracking interactions during 25ns intervals, revealed the key function of Lysine5 in peptide structure and highlighted the importance of realistic model bilayers in forming stable peptide-bilayer interactions.
