چکیده
سرطان پستان از شایعترین سرطانها در بین زنان بوده و اولین علت مرگ و میر ناشی از سرطان در زنان به شمار میرود. ژن کیناز وابسته به سایکلین 4 (CDK4) یکی از مهمترین ژنهای تنظیمکننده ی چرخهی سلولی است و به عنوان انکوژن عمل میکند به طوری که میزان بیان آن در انواع مختلف سرطانها از جمله سرطان پستان افزایش مییابد. شواهد نشان میدهد که اگزوزومهای ترشح شده توسط سلولهای بنیادی مزانشیمی (MSCs) باعث مهار گسترش تومور از طریق انتقال مولکولهای زیستی میشوند. هدف از انجام مطالعه حاضر، تعیین بیان ژن CDK4 و میزان تکثیر و مهاجرت سلولی در ردهی سلولی سرطان پستان 4T1 تحت تأثیر تیمار با اگزوزوم های ترشح شده توسط سلول های بنیادی مزانشیمی می باشد. ردهی سلولی 4T1 سرطان پستان کشت داده شد و با 100 میکروگرم بر میلیلیتر اگزوزومهای MSCs تیمار شد. تغییر بیان ژن CDK4 در سلولهای تیمارشده توسط Real-time PCR ارزیابی شد. همچنین، میزان تکثیر، بقا و مهاجرت سلولهای سرطانی در پی تیمار اگزوزومی سنجیده شد. نتایج نشان داد که تیمار سلولهای سرطان پستان 4T1 با اگزوزومهای مشتق شده از سلولهای بنیادی مزانشیمی باعث کاهش میزان تکثیر و مهاجرت سلولی در مقایسه با گروه کنترل شد. همچنین، کاهش معنیداری در میزان بیان ژن CDK4 به عنوان تنظیم کننده کلیدی چرخه سلولی مشاهده شد. در مجموع این مطالعه پیشنهاد می کند اگزوزوم های مشتق از سلول های بنیادی مزانشیمی اثرات مهاری بر بیان ژن CDK4 در سلول های سرطانی پستان داشته و می توانند سبب کاهش نرخ تکثیر و مهاجرت این سلول ها شوند.

The impact of exosomes derived from bone marrow mesenchymal stem cells on CDK4 gene expression levels, and the proliferation and migration ability of breast cancer cells

Breast cancer (BC) is the most common cancer and the leading cause of death from cancers among women. Cyclin-dependent kinase 4 (CDK4) is one of the most important genes regulating the cell cycle and acts as an oncogene, so that its expression level increases in various types of cancer, including breast cancer. Increasing evidence suggests that exosomes secreted by mesenchymal stem cells (MSCs) inhibit tumor growth by transducing signaling molecules. Therefore, the aim of this study was to determine the expression of CDK4 gene and the rate of cell proliferation and migration in breast cancer cell line 4T1 under the influence of treatment with exosomes secreted by bone marrow mesenchymal stem cells. 4T1 cells were cultured and treated with 100 μg/ml of exosomes. The change of CDK4 gene expression levels was evaluated by real-time PCR. Also, the rate of proliferation and migration rate of cells were measured. Our results showed that treatment of 4T1 breast cancer cells with exosomes derived from bone marrow mesenchymal stem cells decreased the proliferation rate and inhibited cell migration compared to the control group. Also, it led to a significant decrease in CDK4 gene expression as a key regulator of the cell cycle. In conclusion, Exosomes derived from mesenchymal stem cells can significantly reduce the expression of the CDK4 gene and ultimately lead to the reduction of cancer cell proliferation and migration.
