اثر دارویی نانوذرات طلای کوت شده با 6-جینجرول بر سطح هیپوکامپی فاکتور رشد عصبی،فاکتور نوروتروفیک مشتق از مغز،آسیب اکسیداتیو DNA وشاخص های آپوپتوزی در موش های صحرائی متاثر از نانوذرات طلا

چکیده

زمینه و هدف:هدف تعیین اثر نانوذرات طلای کوت شده با ۶-جینجرول برسطح هیپوکامپی فاکتور نوروتروفیک مشتق از مغز، فاکتور رشد عصبی، آسیب اکسیداتیو DNA و شاخصهای آپوپتوزیBax وBcl2 در موش های صحرائی متاثر از نانوذرات طلا می باشد.

• مواد و روش ها: ۲۴سر موش صحرائی نر بالغ نژاد ویستار به 4 گروه 6تایی تقسیم شدند. گروه کنترل به مدت 30 روز به صورت روزدرمیان 5/0میلی لیتر محلول نرمال سالین به صورت داخل صفاقی دریافت و در گروه نانوذرات طلا موش ها یک بار 5/0میلی لیتر محلول نانوذرات طلا ppm۲۰۰ و nm 60 نانومتر به صورت داخل صفاقی دریافت نمودند و پس از یک ماه و به مدت۳۰روز، به صورت روزدرمیان 5/0میلی لیتر سالین تزریق شد. در گروه تجربی1و2: مراحل مشابه گروه نانوذرات طلا بود اما به جای سالین موش ها به صورت روزدرمیان 5/0میلی لیتر نانوذرات طلای کوت شده با 6-جینجرول در گروه یک باغلظتmg/kg 50 و در گروه دو با غلظت mg/kg 100 دریافت نمودند. در پایان دوره درمان سطح هیپوکامپی NGF, BDNF, 8 -OHdG و شاخص های آپوپتوزی Bax و Bcl2توسط روش الایزا مورد سنجش قرار گرفت.

نتایج: در گروه های تجربی درمقایسه با گروه نانوذرات طلا سطح NGF, ، BDNF ،Bcl2 به طور معنی داری افزایش و سطحBax و 8 -OHdGکاهش یافت.(P<0.001) اما در همه موارد به سطح گروه کنترل نرسید.

نتیجه گیری: نانوذرات طلای کوت شده با 6-جینجرول، از طریق کاهش آپوپتوز و کاهش آسیب اکسیداتیو DNA سبب بهبود سطح هیپوکامپی BDNF و NGF ازمغز در موش های صحرائی در معرض نانوذرات طلا می شوند.

چکیده [English]

Background and Aim: The aim was to determine the effects of 6-ginger coated gold nanoparticles on the hippocampal level of brain-derived neurotrophic factor (BDNF), nerve growth factor (NGF), oxidative DNA damage, and apoptotic indices in gold nanoparticles (AuNPs) affected rats.

Materials and Methods: 24 adult male Wistar rats were divided into 4 groups of 6. The animals in the control group received 0.5 ml of saline for 30 days (every other day). In the group of AuNPs, rats received 0.5 ml of gold nanoparticles (200 ppm and 60 nm) dissolved in saline, and after one month 0.5 ml saline for 30 days. In the Experimental Group 1, the same steps were performed in the AuNPs group, but instead of saline, the rats received 0.5 ml of gold nanoparticles coated with 6-gingerol (50 mg / kg). In the Experimental Group 2, similar steps to the AuNPs group were taken, but instead of saline, the animals received 0.5 ml of gold nanoparticles coated with 6-gingerol (100 mg /kg). At the end of the experiments, the level of hippocampal NGF, BDNF, 8-OHdG, and Bax and Bcl2 apoptotic indices were measured by ELISA.

Results: In the experimental groups, compared to AuNPs group, NGF, BDNF, and Bcl2 levels significantly increased, and Bax and 8-OHdG levels decreased significantly (P <0.001). However, all parameters did not reach the control group values.

Conclusion: 6-gingerol coated gold nanoparticles improve the hippocampal level of NGF and BDNF in rat exposed to gold nanoparticles by reducing apoptosis and oxidative DNA damage.
