In recent years, in spite of transformation in canola via Agrobacterium, the efficiency of this method is still low. To consider the effect of plant genotype and cultivation medium composition on transformation and regeneration in canola and the high toxicity of kanamycin for growth of this plant, it is necessary to develop more appropriate and efficient method for transformation of this oily plant. Here we used cotyledon explants and Agrobacterium tumefaciens strains LBA4404, GV3850, GV3101 harboring mutated gene EPSPS (pBI121+EPSPS) for transformation of canola PF7045-91 cultivar. After induction with agrobacterium, explants were placed in different cocultivation medium under dark condition at 25 degree centigrade. In regeneration process, to prevent necrosis of explants, the fixed amount of silver nitrate (5mg/L) was used for blocking ethylene production. The regeneration and selection procedure were started in medium containing silver nitrate without kanamycin and this carried on for one week. Subsequently, the kanamycin was added (5 mg/L) and its concentration was increased up to 15 mg/L in a ten days period. For root induction, the regenerated shoots were transferred to medium containing the MS salt with 15 g/L sucrose and 0.2 mg/L IBA. Transformation was confirmed by molecular analysis. Our data showed that gradually increasing kanamycin concentration and blocking ethylene production could improve the transformation of B. napus.}, keywords = {Brassica napus,Agrobacterium tumefaciens,cocultivation,Control of ethylene production,increase in kanamycin}, title_fa = {بهینه سازی تراریختی در گیاه کلزا (B.napus L. ) با کنترل تولید گاز اتیلن وافزایش تدریجی عامل انتخاب کننده}, abstract_fa = {کلزا (Brassica napus L.) یکی از مهمترین گیاهان روغنی جهان به شمار می رود. علی رغم تولید گیاهان تراریخته کلزا کارایی انتقال به این گیاه پایین می باشد. با توجه به اهمیت تاثیر ژنوتیپ گیاه و ترکیب محیط کشت بر تراریختی و باززایی و همچنین سمیت کانامایسین برای رشد این گیاه، توسعه یک روش مناسب جهت انتقال ژن بسیار ضروری است. در این پژوهش از ریزنمونه های برگهای لپه ای با استفاده از آگروباکتریوم تومی فاشینس سویه های حاوی ژن انول پیروویل شیکیمات 3-فسفات سنتاز ( EPSPS ) برای تراریختی رقم7045-91 PF استفاده شد. ریز نمونه ها پس از تلقیح با اگروباکتریوم محیط های مختلف هم کشتی ، دمای 25 درجه سانتیگراد و تاریکی قرارداده شد. طی باززایی به منظور جلوگیری از نکروز شدن ریز نمونه ها و گزینش موثر از نیترات نقره به عنوان ممانعت کننده تولید اتیلن و افزایش پلکانی غلظت کانامایسین به عنوان ماده انتخاب کننده استفاده شد. برای این منظور ابتدا ریزنمونه ها در محیط باززایی حاوی نیترات نقره و بدون حضور کانامایسین نگهداری شدند. پس از آن با حفظ غلظت ثابتی از نیترات نقره (5 میلی گرم بر لیتر)، کانامایسین به غلظت 15میلی گرم بر لیتر افزایش داده شد. نوساقه های باززا شده به منظور القا ریشه زایی به محیط MS حاوی 15گرم بر لیتر ساکارز و غلظت 2/0 میلی گرم بر لیتر از IBA منتقل شدند. انتقال موثر ژن به گیاهان با آزمون های مولکولی تایید شد. در این بررسی مشخص شد که با افزایش تدریجی عامل انتخابی، درصد تراریختی نیز افزایش می یابد.}, keywords_fa = {Brassica napus,Agrobacterium tumefaciens,cocultivation,Control of ethylene production,increase in kanamycin}, url = {https://cell.ijbio.ir/article_519.html}, eprint = {https://cell.ijbio.ir/article_519_d743619ac0964238cc9db5986f27ff22.pdf} }
