They increase soil productivity by affecting its physicochemical properties. Increasing the application of chemical fertilizers is harmful. Therefore, it has become necessary to look for alternative renewable resources to meet at least a part of the demand for crops. In this study, aimed at increasing the efficiency of cyanobacterial biofertilizer, the effects of nitrogen, phosphorus and potassium elements in the form of urea, superphosphate and potassium chloride fertilizers were evaluated on nitrogen fixation activity measured by gas chromatography, nifH gene expression and the growth of cyanobacterium Aliinistoc sp. In general, the growth and nitrogen fixation activity of the sample decreased as the concentration of nitrogen and phosphorus sources increased. The most effect of nitrogen source was on nitrogenase activity, and at the end of the experimental period compared to the beginning (before treatment), the amount of nitrogenase activity decreased 50 and 100% in the presence of 10 and 100 mg/L urea, respectively. Phosphorus had a significant effect on the growth of the sample, and at the highest concentration of superphosphate (500 mg/L), growth declined about 80%. On the other hand, potassium especially at the lowest concentration (125 mg/L), significantly increased growth by about 20% and doubled nitrogenase activity. In addition, the evaluation of nifH gene expression confirmed the pattern of nitrogenase activity. In general, the results of this study indicate that the dosage of elements used in combination with cyanobacterial biofertilizers should be considered.}, keywords = {Cyanobacteria,Biofertilizer,nifH gene,Nitrogen fixation,Real-time RT-PCR}, title_fa = {بررسی فعالیت تثبیت نیتروژن، بیان ژن nifH و رشد در سیانوباکتر Aliinostoc sp.، تحت تاثیرمنابع نیتروژن، فسفر و پتاسیم}, abstract_fa = {سیانوباکترها قدیمی‌ترین فتواتوتروف‌های اکسیژنی با توزیع اکولوژیکی گسترده‌ بر روی زمین هستند. آنها با اثر بر خصوصیات فیزیکوشیمیایی خاک، موجب افزایش حاصلخیزی آن می‌شوند. به دلیل مضرات افزایش مصرف کودهای شیمیایی، جستجو برای منابع زیستی که حداقل قسمتی از نیاز محصول را فراهم کند، ضروری است. در این مطالعه با هدف افزایش کارایی کود زیستی سیانوباکتریایی، تاثیر عناصر نیتروژن، فسفر و پتاسیم در قالب کودهای اوره، سوپرفسفات و کلرید پتاسیم، بر فعالیت تثبیت نیتروژن اندازه‌گیری شده به وسیله کروماتوگرافی گازی، بیان ژن nifH و رشد سیانوباکتر Aliinistoc sp.، بررسی شد. به طور کلی افزایش غلظت منابع نیتروژن و فسفر، رشد و فعالیت تثبیت نیتروژن را کاهش دادند. بیشترین اثر منبع نیتروژن بر میزان فعالیت نیتروژناری بود، به طوری‌که در انتهای دوره آزمایشی در مقایسه با ابتدای آن (پیش از تیمار)، میزان فعالیت نیتروژنازی در حضور 10 و 100 میلی‌گرم در لیتر اوره، به ترتیب 50 و 100 درصد کاهش یافت. فسفر اثر قابل توجهی بر رشد نمونه نشان داد و در بیشترین غلظت مورد استفاده از سوپرفسفات (500 میلی‌گرم در لیتر)، کاهش رشد حدود 80 درصدی مشاهده گردید. از طرف دیگر، پتاسیم به ویژه در کمترین غلظت مورد استفاده (125 میلی‌گرم در لیتر)، موجب افزایش قابل توجه حدود 20 درصدی رشد و افزایش دو برابری فعالیت نیتروژنازی شد. علاوه براین، ارزیابی بیان ژن nifH الگوی فعالیت نیتروژنازی را تایید نمود. در مجموع نتایج این مطالعه بیانگر لزوم توجه به مقدار عناصر مورد استفاده به همراه کود زیستی سیانوباکتریایی است.}, keywords_fa = {Cyanobacteria,Biofertilizer,nifH gene,Nitrogen fixation,Real-time RT-PCR}, url = {https://cell.ijbio.ir/article_2147.html}, eprint = {} }
