Cancer stem cells secret different materials such as protease enzymes. Interstitial collagenase or fibroblast collagenase is a member from large family of protease enzymes destroying basic membrane and extracellular matrix. Therefore, it is not only facilitates progression of cancer cells but also having key role in cancer cell viability through releasing of growth and angiogenesis factors. Insertion of a guanine nucleotide at the position of -1607 introduces a binding site for ETS transcriptional factor in the promoter of the gene. Binding ETS to the 2G polymorphism allele would be increased expression of fibroblast collagenase gene which it could be progressed releasing of cancer stem cells, and therefore, it could be facilitated metastasis. Goal of this study is valuation of association between the 2G/1G polymorphism with progression and metastasis of breast cancer, and also patient viability. Blood samples of 200 cases and 100 controls were collected from two capital cities, Tehran and Isfahan; equally. Samples were genotyped using PCR-RFLP method. Statistical analyses show that 2G/2G genotype frequency is much more in cases rather than controls (29.5% compare with 23%). Patients were divided into two groups of with metastatic activity (M+) and without metastatic activity (M-). The 2G/2G genotype was more frequent in M+ group compared with control group (OR = 2.03, CI95% = 1.05–3.94). In next step, patients without metastasis were followed up for 32 months. Three years total viability analyses in patients who carrying the 2G/2G genotype in comparing with other patients showed no significantly difference. However rate of cancer special viability and viability without disease in the two groups were statistically significant. In conclusion, to our knowledge, the present epidemiological study for the first time indicates that the 2G/2G genotype polymorphism could be a facilitated factor for progression and metastasis of breast cancer and causing of viability reduction in patients.}, keywords = {Cancer stem cells,Collagenase fibroblast,Viability rate,Metastasis,Breast cancer}, title_fa = {نقش سلولهای بنیادی سرطانی در افزایش ریسک ابتلاء به متاستاز و کاهش میزان بقای افراد مبتلا به سرطان پستان}, abstract_fa = {امروزه سلولهای بنیادی سرطانی به عنوان یکی از عوامل اصلی و منشاء متاستاز مطرح می باشند. سلولهای بنیادی سرطانی مواد متعددی از جمله آنزیمهای پروتئازی را از خود ترشح می کنند. آنزیم کلاژناز بینابینی یا کلاژناز فیبروبلاستی عضوی از خانواده بزرگ آنزیمهای پروتئازی مذکور می باشد که با تخریب غشای پایه و ماتریکس خارج سلولی نه تنها گسترش سلولهای سرطانی را تسهیل می نماید بلکه با رهایش فاکتورهای رشد و فاکتورهای رگزایی در بقاء و تغذیه سلولهای سرطانی نیز نقش کلیدی ایفا می کند. دخول یک باز گوانین در موقعیت 1607- پروموتور ژن کلاژناز فیبروبلاستی یک جایگاه اتصال برای اعضای خانواده ETS از فاکتورهای رونویسی ایجاد می کند. جذب عوامل الگوبرداری در اثر این پلی مورفیسم تک نوکلئوتیدی (G2) بیان ژن کلاژناز فیبروبلاستی را به طور محسوسی افزایش می دهد که این شرایط می تواند گسترش سلولهای توموری و تهاجم آنها را آسان تر سازد. هدف از این مطالعه ارزیابی اثر پلی مورفیسم مذکور بر گسترش و متاستاز سرطان پستان و میزان بقای کل، بقای ویژه سرطان و بقای عاری از بیماری افراد مبتلا به سرطان پستان است. بدین منظور، نمونه خون 200 خانم مبتلا به سرطان پستان و 100 نمونه کنترل به نسبت مساوی از دو شهر تهران و اصفهان جمع آوری گردید و با روش PCR-RFLP تعیین ژنوتیپ گردید. محاسبات آماری نشان دادند که فراوانی ژنوتیپ G2/G2 در بین افراد بیمار نسبت به افراد کنترل بیشتر است (5/29 درصد در برابر 23 درصد). بیماران در دو دسته متاستازی (M+) و غیر متاستازی (M-) طبقه بندی شدند. توزیع ژنوتیپی G2/G2 در گروه متاستازی نسبت به گروه کنترل تفاوت آماری معنی داری نشان داد (94/3-05/1=95% CI و 03/2=OR). در مرحله بعد بیماران غیر متاستازی به مدت میانگین 32 ماه تحت نظارت و پیگیری قرار گرفتند. آنالیز بقای کلی 3 ساله در افراد بیمار واجد ژنوتیپ G2/G2 در مقایسه با افراد واجد ژنوتیپهای دیگراز لحاظ آماری معنی دار نبود (تست لاگ-رانک: 12/0=P، 34/2=χ²). اما میزان بقای ویژه سرطان و بقای عاری از بیماری در بین دو گروه مذکور تفاوت آماری معنی داری نشان داد (بقای ویژه سرطان، تست لاگ-رانک:  01/0=P، 45/5 = χ² ، بقای عاری از سرطان، تست لاگ-رانک: 002/0=P، 99/8 =χ²). در مجموع، مطالعه اپیدمیولوژی حاضر برای اولین بار نشان داد که ژنوتیپ G2/G2 به عنوان یک فاکتور تسهیل کننده، با گسترش و متاستاز سرطان پستان مرتبط بوده و سبب افزایش ریسک ابتلاء به متاستاز و کاهش میزان بقاء در افراد واجد ژنوتیپ G2/G2 می شود.}, keywords_fa = {سلولهای بنیادی سرطانی,کلاژناز فیبروبلاستی,میزان بقاء,متاستاز,سرطان پستان}, url = {https://cell.ijbio.ir/article_209.html}, eprint = {https://cell.ijbio.ir/article_209_4e0a274cfe1c177eb1d1b7e4aaf55dc4.pdf} }
