Due to its antioxidant properties, boswellia is effective in reducing the progression of neurological diseases. In this work, effects of Acetyl-11-keto-β-boswellic acid derivatives on the monomer and dimer structure of Aβ (1-40) and Aβ (1-42) peptides are monitored by using molecular dynamics simulation. The results showed that in the presence of ligand, the flexibility of the C-terminal residues of Aβ (1-40) is higher. The ligand also reduces the amount of peptide sheets. By calculating the conformation factor, the afinity of the ligand to interact with each of the peptide residues was calculated. The results of this calculation showed that the residues that tend to be binded have less flexibility. By calculating the free binding energy, the binding site of the ligands was determined.}, keywords = {Alzheimer,peptide,Aggregation,ligand}, title_fa = {تاثیر مشتق های بوسولیک اسید 3-O-Acetyl-11-keto-β-boswellic acid (AKBA) روی ساختار مونومر و دیمر Aβ(1-40) و Aβ(1-42) : نگرش شبیه سازی دینامیک مولکولی}, abstract_fa = {دو پپتید Aβ (1-40) و Aβ (1-42) در بیماری آلزایمر موثر هستند که به صورت تجمعات فیبریلی خارج سلول عصبی، باعث ایجاد بیماری می شوند. یکی از روش های مهار پیشرفت بیماری آلزایمر، تجویز داروهایی است که باعث کاهش این تجمعات شود. از جمله این داروها، داروهایی با منشا گیاهی هستند که عوارض جانبی کمتری هم دارند. گیاه بوسولیا (Boswellia) با خواص آنتی اکسیدانی بالا، اثرات درمانی در برابر بیماری های مختلف از جمله بیماری های عصبی دارد. در این تحقیق، برهم کنش یک مشتق بوسولیک اسید با نام 3-O-acetyl-11-keto-b-Boswellic acid (AKBA) به عنوان لیگاند با پپتیدهای Aβ (1-40) و Aβ (1-42) به صورت مونومر و دیمر به روش شبیه سازی دینامیک مولکولی بررسی شد. نتایج آنالیزهای مختلف نشان دادند بیشترین میزان انعطاف دنباله ها در دنباله های C ترمینال پپتید دوم Aβ (1-40) و در حضور لیگاند است. همچنین، لیگاند تاثیر بازدارندگی و کاهش تشکیل ساختار صفحه را بر دنباله های پپتید Aβ (1-40) نشان می دهد. افینیته اتصال لیگاند به دنباله هایی از پپتیدها که بیشترین برخورد با لیگاند را داشتند، با استفاده از معیار فاکتور صورتبندی محاسبه شد و مشخص شد این دنباله ها انعطاف پذیری کمتر و در نتیجه تمایل بیشتری به پیوند با لیگاند دارند. در نهایت، جایگاه های اتصال از طریق محاسبه انرژی اتصال دنباله های مذکور معرفی شد که نتایج حاصل از این شبیه سازی دینامیک مولکولی تطابق خوبی با شواهد تجربی مربوطه داشت.}, keywords_fa = {آلزایمر,پپتید,تجمع,لیگاند}, url = {https://cell.ijbio.ir/article_2067.html}, eprint = {https://cell.ijbio.ir/article_2067_091cf57e40f4b5a77c066f3a46c6dd27.pdf} }
