Production of recombinant proteins in E. coli has particular disadvantages, which are primarily inclusion body formation and lack of disulfide bond formation in the cytoplasm. We therefore aimed to use the modified signal peptide of the Iranian native Bacillus licheniformis alpha-amylase to target the expressed recombinant β-NGF to the periplasm as an effective strategy to produce correctly-folded β-NGF.For this purpose, the human β-NGF gene with the signal sequence of the Iranian native Bacillus licheniformis alpha-amylase was cloned into a pET21a (+) vector and then the recombinant vector was transformed to BL21 (DE3) and BL21 (DE3) plysS strains. Protein expression was induced by 1mM IPTG in strains with the recombinant vector and the positive control strain containing the pET39b (+) vector with the DsbA signal peptide. Cytoplasmic and periplasmic expression of β-NGF was confirmed by SDS-PAGE and dot-blot assays. Finally, the expressed periplasmic proteins were purified using affinity chromatography and this purification was confirmed by using SDS-PAGE and Western blot assays.The results show the modified signal peptide of the Iranian native Bacillus licheniformis alpha-amylase is more effective for the secretory expression and directing β-NGF to the periplasmic space than the DsbA signal peptide. Also, these results indicate that the BL21 (DE3) plysS strain is an appropriate host for periplasmic expression of β-NGF.}, keywords = {Nerve growth factor,Recombinant protein,signal sequence,periplasmic expression}, title_fa = {کلون سازی و بیان ترشحی فاکتور رشد بتا-NGF انسانی با استفاده از پپتید نشانه ی اصلاح شده ی آنزیم آلفا آمیلاز باسیلوس لیکنی فورمیس بومی ایران}, abstract_fa = {فاکتور رشد عصبی(NGF) عضوی شناخته شده از خانواده ی پروتئین های نوروتروفین است که در درمان بیماری هایی مانند مولتی پل اسکلروزیس و آلزایمر بکار گرفته می شود. تولید پروتئین نوترکیب در E. coli دارای معایب خاصی است که شکل گیری اینکلوژن بادی و عدم شکل گیری پیوندهای دی سولفیدی درسیتوپلاسم در درجه اول قرار دارند .بنابراین ما به منظور هدایت β-NGF نوترکیب تولید شده به سمت پری پلاسم، استفاده از پپتید نشانه ی اصلاح شده آنزیم آلفا آمیلاز باسیلوس لیکنی فورمیس بومی ایران را به عنوان یک رویکرد مؤثر برای تولید β-NGF با تا خوردگی صحیح، مورد هدف قرار دادیم. به همین منظور ژن β- NGF انسانی به همراه توالی پپتید نشانه مذکور در وکتور pET21a(+)کلون شدند و سپس وکتور نوترکیب به سویه های BL21(DE3) و BL21(DE3)plysS انتقال داده شد. بیان پروتئین درسویه های دارای وکتور نوترکیب و سویه ی کنترل مثبت حاوی وکتور pET39b(+) دارای پپتید نشانه DsbA با استفاده از IPTG ، 1 میلی مولار القا گردید. بیان سیتوپلاسمی وپری پلاسمی β-NGF با تکنیک های SDS-PAGE و سنجش دات بلات تأیید شد. در نهایت، پروتئین های پری پلاسمی بیان شده با استفاده از کروماتوگرافی تمایلی تخلیص وصحت تخلیص با روش SDS-PAGE و وسترن بلات تأیید شد. نتایج نشان می دهد که در تولید ترشحی و هدایت پروتئین β-NGF به فضای پری پلاسمی توالی پپتید نشانه ی اصلاح شده ی آنزیم آلفا آمیلاز باسیلوس لیکنی فورمیس بومی ایران نسبت به پپتید نشانه DsbA عملکرد بهتری داشته و سویه ی BL21(DE3)plysS میزبان مناسبتری می باشد.}, keywords_fa = {فاکتور رشد عصبی,پروتئین نوترکیب,توالی نشانه,بیان پری پلاسمی}, url = {https://cell.ijbio.ir/article_1604.html}, eprint = {https://cell.ijbio.ir/article_1604_51f44906f58c5d1980635a28c4ba6759.pdf} }
