In the present study, Keratinase production was examined and optimized in FUM120 Keratinolytic strain isolated from a poultry farm in Mashhad. Using of Azokeratin substrate, it was determined that the isolate produces the most in the 72nd hours of its growth with the amount of 49.965 U/ml. Then, In order to increase the production of keratinase, the single-variable optimization of parameters, temperature, pH, substrate concentration, extra carbon and nitrogen sources, inoculum size and aeration rates were done and it was determined that pH:10.5 and 0.6% concentration of ammonium chloride, as an additional source of nitrogen, at 37 °C had a significant effect on keratinolytic activity and increased the enzyme production by more than 2 times. In addition, the isolate was able to produce the high level of enzyme in the presence of 50% aeration without the addition of carbon source. Finally, enzyme production reached to 117/45 U/ml and the enzyme unit showed an increase of 2.34 fold compared to the beginning of the optimization. The morphological studies and the gene sequence of 16SrRNA determined that FUM120 belongs to the genus Bacillus and shows 100 % similarity to Bacillus pumilus ATCC 7061.}, keywords = {keratin,feather,azokeratin,Bacillus pumillus,keratinolytic activity}, title_fa = {بهینه سازی تولید آنزیم کراتیناز توسط جدایه بومی Bacillus sp FUM 120 به روش تک متغیره}, abstract_fa = {امروزه با توجه به کاربرد گسترده آنزیم کراتیناز در صنایع مختلف از جمله خوراک دام، صنعت چرم، تصفیه فاضلاب، پزشکی و داروسازی تقاضا برای این آنزیم با کاربرد صنعتی رو به افزایش است. در پژوهش حاضر تولید آنزیم کراتیناز در جدایه FUM120 کراتینولیتیک، جدا شده از مرغداری های اطراف مشهد مورد بررسی و بهینه‌سازی قرارگرفت. با استفاده از سوبسترای آزوکراتین، مشخص گردید که این باکتری در ساعت 72 رشد خود با U/ml 965/49 بیشترین میزان تولید خود را دارا می‌باشد. سپس بهینه سازی تک متغیره پارامترهای دما، pH، غلظت سوبسترا، منابع کربن و نیتروژن اضافه، میزان تلقیح و هوادهی به منظور افزایش تولید آنزیم کراتیناز انجام گرفت و مشخص گردید 5/10:pH و غلظت 6/0 درصد آمونیوم کلرید به‌عنوان منبع نیتروژن اضافه در دمای C◦ 37 تاثیر قابل توجهی در افزایش فعالیت کراتینولیتیکی دارند و باعث افزایش بیش از 2 برابر تولید آنزیم گردیدند. همچنین باکتری بدون نیاز به منبع کربن اضافه با میزان هوادهی 50٪ قادر به تولید مقادیر بالایی از آنزیم بود. در نهایت تولید آنزیم به U/ml 45/117 واحد آنزیمی رسید و افزایش 34/2 برابری را نسبت به ابتدای بهینه سازی نشان داد. با بررسی خصوصیات مورفولوژیکی و توالی ژن 16S rRNA مشخص گردید، جدایه FUM120 متعلق به جنس باسیلوس می باشد و 100٪ مشابه سویه Bacillus pumillus ATCC 7061 میباشد.}, keywords_fa = {keratin,feather,azokeratin,Bacillus pumillus,keratinolytic activity}, url = {https://cell.ijbio.ir/article_1594.html}, eprint = {https://cell.ijbio.ir/article_1594_420ca87de5ef5625d74b90c69f8bcdb5.pdf} }
