بررسی نقش آمینواسیدهای اطراف توالی جایگاه فعال در فعالیت آنزیم کیتیناز باکتری Serratia Marcescens B4A
کیتینازها یکی از آنزیمهای صنعتی مهم محسوب میشوند که از اهمیت بسزایی در خصوص تجزیه مواد کیتینی، پاکسازی محیط زیست و کنترل قارچهای پاتوژن گیاهی و آفات دارند که با شکستن اتصالات گلیکوزیدی -1,4 سبب تجزیه کیتین میشوند. این آنزیمها در ساختار خود دارای دمین کاتالیتیکی 8 (β/α) حاوی توالیهای حفاظت شدهای به نام موتیف DXDXE روی رشته 4β هستند که آسپارتات و گلوتامات موجود در این توالی و همچنین آمینواسیدهای موجود در اطراف این توالی نقش اصلی در فرایند هیدرولیز پیوند گلیکوزیدی بازی میکنند. بررسی آمینو اسیدهای نزدیک به جایگاه فعال می تواند به درک نقش آنها در فرایند کاتالیز کمک کند. در این پروژه به منظور بررسی نقش گلیسین 191 و سرین 390 موجود در اطراف توالی حفاظت شده در فرایند کاتالیتیکی کیتیناز Serratia marcescens B4A، این دو آمینواسید جهش داده شدند و پس از همسانهسازی در حامل بیانی و بررسی بیان آن با SDS-PAGE، اثر این دو جهش بر روی فعالیت آنزیم مورد بررسی قرار گرفت.

تولید ترکیبات فنلی درکشت ریشههای مویین گیاه تربچه (Raphanus sativus L.)
اگروباکتریوم رایزوژنز سبب ایجاد بیماری ریشه مویین در گیاهان میشود. ریشههای مویین تولید شده توسط آلودگی با این باکتری، با نرخ بالای رشد و ثبات ژنتیکی مشخص میشوند. ایجاد ریشه مویین ممکن است سبب تغییراتی در تولید ترکیبات ثانویه گیاه شود. ترکیبات فنلی (فلاونوئیدها، تانن ها و آنتوسیانین ها) از جمله متابولیتهای هستند که جزء آنتی اکسیدان های طبیعی به شمار آمده و در گیاه تربچه به مقدار فراوان حضور دارند. در این مطالعه جهت ایجاد ریشههای مویین، تراریختی ریزنمونه های برگی گیاه تربچه با استفاده از دو سویه اگروباکتریوم رایزوژنز (A4 و GUS+) انجام شد. ریشههای مویین ایجاد شده با استفاده از آزمون PCR مورد تایید قرار گرفتند. همچنین میزان ترکیبات فنلی در ریشههای مویین مورد سنجش قرار گرفت. صحت تکثیر قطعات 780 جفت بازی برای ژن rolB و 320 جفت بازی برای ژن GUS در ریشهها بیانگر تراریخت بودن آنها بود. نتایج مقایسه میانگینها نشان داد که میزان ترکیبات فنلی در ریشههای مویین به طور معنی داری نسبت به ریشههای عادی افزایش یافت. به نظر میرسد افزایش ترکیبات فنلی در ریشههای مویین نتیجه ورود T-DNA اگروباکتریوم رایزوژنز و برانگیختن پاسخ دفاعی گیاه باشد.

بهینه سازی تولید لیپاز مخمر یارروویا لیپولیتیکا با روش طراحی آزمایش تاگوچی
لیپاز (EC 3.1.1.3) یک دسته بسیار مهم از آنزیم های صنعتی است. مخمر غیرمعمول یارروویا لیپولیتیکا برای انسان غیر بیماریزا و برای چند فرآیند صنعتی ایمن شناخته شده است. این مخمر می تواند انواع مختلفی از لیپاز را تولید کند. تولید لیپاز خارج سلولی به ترکیب محیط کشت و شرایط محیطی وابسته است. روغن زیتون به عنوان منبع کربن، عصاره مخمر و تریپتون به عنوان منابع نیتروژن و pH در سطوح مختلف برای بهینه سازی تولید لیپاز در سویه جهش یافته مخمر Yarrowia lipolytica FDY1390 توسط روش طراحی آزمایش تاگوچی مورد بررسی قرار گرفتند. نرم افزار Qualitek-4 برای طراحی آزمایش به روش تاگوچی استفاده شد. بیشترین تولید آنزیم لیپاز (340 واحد در میلی لیتر) در محیط کشت حاوی 20 گرم در لیتر روغن زیتون ، 15 گرم در لیتر عصاره مخمر و تریپتون و pH برابر 4 پس از 72 ساعت از فرآیند تخمیر بدست آمد. مطالعه و بهینه سازی تولید لیپاز می تواند سبب کاهش هزینه تولید و استفاده صنایع مختلف از این آنزیم می شود.

مطالعه پایداری ساختاری آنزیم پپسین در حضور نانوذرات اکسیدروی و اکسیدآهن
آنزیم پپسین خوک A (EC 3.4.23.1) متعلق به خانواده آسپارتیک پروتئازها است، و به صورت زیموژن که پپسینوژن نام دارد ترشح می شود، فعالشدن پپسینوژن در مقادیر pH بین 1 و 4 اتفاق می افتد. پپسین شامل یک صورتبندی دو لوبی با دو رزیدو آسپارتیک کاتالیتیکی(32Asp و215Asp) که در دو طرف جایگاهفعال می باشد و ساختار آن غالباً صفحه بتا و رندوم کویل با مارپیچ آلفا محدود میباشد. پپسین پیوندهای پپتیدی بین آمینواسیدهای هیدروفوبیک و ترجیحاً آروماتیک از قبیل تریپتوفان، فنیلآلانین و تیروزین را هیدرولیز میکند. با توجه به اهمیت آنزیم پپسین به عنوان یک آنزیم صنعتی در صنایع غذایی و کاربردهای نانوذرات در صنعت ما اثر نانوذرات اکسید مس و اکسید روی را بر روی پایداری ساختاری پپسین بررسی کردیم. این مطالعه با استفاده از بافر گلایسین-اسیدکلریدریک (0.1 مولار) با 2=pH و در دامنه دمایی 303 تا 363 درجه کلوین انجام شد. پایداری پپسین در حضور نانوذرات اکسید روی و اکسید آهن تغییر نکرد. قدرت برهم کنش الکتروستاتیک و هیدروژنی بین آنزیم پپسین و نانوذرات اکسیدروی و اکسیدآهن در دگرگون شدن پپسین کم بوده است.

جداسازی، همسانه سازی و بیان ژن زیرواحد آلفا فیکوسیانین در سیستم بیانی اشرشیاکلی
فیکوسیانین رنگدانه آبی رنگی است که در دو گروه از جلبک ها و سیانوباکترها ازجمله Spirulina وجود دارد. این رنگدانه دارای فعالیت های متنوع زیستی است و کاربردهای گوناگونی در صنعت غذایی، آرایشی و دارویی دارد. پیشرفت های زیادی جهت تولید فیکوسیانین در مقیاس بالا انجام شده است، اما اغلب روش ها مشکل و با هزینه های بالایی قابل انجام هستند؛ درصورتی که همسانه سازی و بیان فیکوسیانین به صورت نوترکیب روشی ارزان است و خالص سازی آن آسان تر انجام می پذیرد. از این رو هدف از این تحقیق، جداسازی و همسانه سازی ژن زیرواحد آلفا فیکوسیانین در ناقل بیانی و تولید این پروتئین نوترکیب در باکتری اشرشیاکلی بود تا زمینه تولید فیکوسیانین به صورت صنعتی فراهم گردد. در این پژوهش ژنوم سیانوباکتر Spirulina platensisاستخراج شد و به عنوان الگو در PCR مورد استفاده قرارگرفت. ژن زیرواحدآلفا فیکوسیانین تکثیریافته با آغازگرهای طراحی شده، در ناقل بیانی +pET-43.1a با استفاده از آنزیم های برشی NdeIوNotI کلون گردید. همسانه سازی ژن زیرواحد آلفا در ناقل بیانی با استفاده از Colony PCR، هضم آنزیمی و تعیین توالی تایید شد. بیان ژن با استفاده از آنالیز SDS-PAGE 12/5٪ تا 8ساعت پس از القا با IPTG مورد بررسی قرارگرفت. در بررسی SDS-PAGE، بیان قوی ژن زیرواحدآلفا فیکوسیانین در سیستم بیانی اشرشیاکلی تائید شد. بیان بالای ژن زیرواحدآلفا فیکوسیانین نشان داد که باکتری اشرشیاکلی می تواند به عنوان میزبان مناسب، جهت تولید فیکوسیانین نوترکیب مورد استفاده قرارگیرد. همچنین این تحقیق زمینه تولید فیکوسیانین نوترکیب در آینده را با صرف هزینه های پایین تر فراهم آورد.

تنوع فیلوژنتیکی باکتریهای قابل کشت دریاچه بزنگان و نقش اکولوژی آنها
باکتریها فراوان ترین و غنی ترین گروه از موجودات زنده اند که در بسیاری از فرایندهای مهم اکوسیستم نقش دارند. با وجود اهمیت اکولوژیکی آنها اطلاعات در مورد تنوع زیستی به ویژه در محیط های آبی ناچیز می باشد. ایران دارای دریاچه های متنوعی در نقاط مختلف می باشد که هر کدام به دلیل اقلیم ویژه ای که در آن قرار دارند، از نظر اکولوژی دارای ویژگیهای منحصر به فردی می باشد. دریاچه بزنگان تنها دریاچه مهم و طبیعی استان خراسان رضوی است که جزء آبهای کم شور محسوب می شود و تاکنون پژوهشی درباره میکروارگانیسم های آن انجام نشده است. پژوهش حاضر به بررسی تنوع فیلوژنتیکی باکتریهای قابل کشت موجود در دریاچه بزنگان پرداخته است. نمونه برداری در آبان 1391 صورت گرفت و غربالگری باکتریها منجر به جداسازی 51 باکتری گرم منفی و 15 باکتری گرم مثبت گردید. 30 جدایه به منظور شناسایی مولکولی با استفاده از ژنS rRNA 16، بررسی ویژگیهای مورفولوژی، بیوشیمیایی و آنزیم های هیدرولازی انتخاب شدند. سویه های شناسایی شده متعلق به گروههای beta-Gamma-Proteobacteria و Bacteroidetes و Firmicutes می باشند. تنوع جنس های متعلق به گرم منفی بیشتر بوده و شامل Pseudomonas، Xanthomonas، Luteibacter، Varivorax، Collimonas و Flavobacterium می باشند. در حالی که باکتریهای گرم مثبت از تنوع و تعداد کمتری برخوردار بوده و شامل جنس های Bacillus، Fictibacillus، Staphylococcus و Paenibacillus می باشند. بیشترین فراوانی مربوط به جنس Pseudomonas می باشد. بررسی آنزیم های هیدرولازی در بین سویه های گرم مثبت و گرم منفی تفاوت معنی داری را نشان نمی دهد.

تجزیه و تحلیل کاریوتیپی ژنوتیپهای بومادران با استفاده از روشهای آماری چند متغیره
به منظور مطالعه سیتوژنتیکی و تعیین سهم هر یک از صفات کاریوتیپی در ایجاد تنوع شش ژنوتیپ از دو گونه بومادران A. millefolium و A. santolina مورد بررسی قرار گرفت. برای تهیه نمونه کروموزومی مناسب 5/0 سانتی متر نوک ریشه جدا و پس از پیش تیمار تثبیت گردید. بعد از هیدرولیز، اسکواش و در نهایت عکسبرداری و تهیه کاریوتیپ صورت گرفت. عدد پایه کروموزومی در تمام ژنوتیپها x=9 بود. ژنوتیپهای الیگودرز، اردبیل، مشکین شهر دیپلوئید و اراک، ایلام و استهبان تتراپلوئید بودند. براساس جدول دو طرفه استبینز اراک و ایلام در کلاس 1A، اردبیل و مشکین شهر در کلاس 2A و الیگودرز و استهبان در کلاس 1B قرار گرفتند. مقایسه میانگین ها نشان داد که از نظر S، L و T استهبان دارای کمتربن و الیگودرز دارای بیشترین مقدار میباشد. در تجزیه به عاملها دو عامل در مجموع بیش از 92/90 درصد از کل تغییرات دادهها را توجیه کردند. عامل اول با توجیه 65/46 درصد از تغییرات شامل ضرایب عاملی مثبت و معنی دار برای صفات L/S و L-S و ضرایب عاملی منفی و معنی دار برای صفات %F و S/L بود؛ لذا عامل نسبت بازوهای کروموزومی و شاخصهازیوارا نامگذاری شد. عامل دوم با توجیه 27/44 درصد ازتغییرات، دارای ضرایب عاملی مثبت و معنی دار برای صفات S، L، T و %RL بود؛ لذا عامل اندازه کروموزوم یا ژنوم نامیده شد. تجزیه خوشهای ژنوتیپها را در دو خوشه گروه بندی نمود. در تجزیههای آماری از نرم افزارهای Excel (2007)، Photoshop (Adobe Photoshop CS2) و SPSS (PASW Statistics18) استفاده شد.

جداسازی و بررسی ویژگیهای باسیلوس MF3 تجزیه کننده سیانید در شرایط قلیایی
سیانید ترکیبی خطرناک برای موجودات زنده است. ترکیبات سیانیدی به طور گسترده در صنایع مختلف مورد استفاده قرار گرفته و موجب آلودگی محیط زیست میشود. تجزیه زیستی بهترین روش برای از بین بردن سیانید در پساب صنایع و معادن میباشد. جداسازی باکتریهای تجزیه کننده سیانید در شرایط قلیایی از خاک آلوده و مطالعه توانایی آنها در تجزیه سیانید، از اهداف این پژوهش بود. نمونههای خاک آلوده به سیانید در محیط کشت معدنی حداقل و حاوی 3/4 میلی مولار پتاسیم سیانید، غنیسازی شد. سپس تجزیه سیانید و تولید آمونیاک در محیط رشد به روش پیکریک اسید و نسلر، سنجیده شد. همچنین توانایی باکتری جداشده در استفاده از ترکیبات مختلف سیانیدی بررسی شد. با روش غنی سازی متوالی، جدایه باکتریایی با توانایی تجزیه سیانید در شرایط قلیایی، جداسازی شد و MF3 نامگذاری شد. جدایه MF3 توانست سیانید موجود در محیط رشد را در شرایط قلیایی و پس از 36 ساعت تجزیه کند. نتایج نشان داد که کاهش غلظت سیانید با افزایش غلظت آمونیاک در محیط رشد و نیز رشد MF3، ارتباط مستقیم داشت. همچنین باکتری جداشده، توانایی استفاده از ترکیبات مختلف سیانیدی به عنوان تنها منبع کربن و نیتروژن را نشان داد. بررسی توالی ژن 16S rDNA و رسم درخت فیلوژنی نشان داد که جدایه MF3 دارای 99 درصد هومولوژی با باسیلوس سافِنسیس (Bacillus safensis) بود. نتایج تحقیق حاضر نشان داد که جدایه MF3، گزینه مناسبی برای تجزیه زیستی سیانید در شرایط قلیایی است. این باکتری میتواند برای حذف سیانید از پساب صنایع و مکانهای آلوده، معرفی شود.

تنوع آللی ژن کالپاستاتین در گوسفند سنجابی
تولید گوشت ترد که مطلوب مصرف کنندگان باشد یکی از مسائل مهم در صنعت پرورش گوسفند است. لذا، مطالعه سازکارهای بیوشیمیایی تجزیه ماهیچه در سطح مولکولی ضروری است. کالپاستاتین یک مهار کننده داخلی است و نقش اصلی را در تنظیم فعالیت کالپاین در سلولها ایفاء می کند. گوسفند سنجابی یک حیوان مهم تولید کننده گوشت در استان کرمانشاه است که با نشانگرهای مولکولی، به ویژه از نظر ژن کالپاستاتین مورد مطالعه قرار نگرفته است. بنابراین، هدف این تحقیق تعیین تنوع ژنتیکی گوسفندان سنجابی از نظر ژن کالپاستاتین و مقایسه این نژاد با نژادهای دیگر بود. برای انجام این مطالعه یک قطعه 622 جفت بازی از این ژن از روی 142 نمونه DNA گوسفند سنجابی با واکنش زنجیره ای پلیمراز (PCR) تکثیر شد. محصولات PCR با روش RFLP و با استفاده از دو آنزیم MspI و NcoI هضم و سه ژنوتیپ MM، MN و NN مشاهده شدند. فراوانی ژنوتیپهای MM، MN و NN به ترتیب 67/0، 25/0 و 08/0، فراوانی ژنهای M و N به ترتیب 79/0 و 21/0 و میانگین شاخصهای شانون و نئی به ترتیب 51/0 و 33/0 به دست آمد و جمعیت برای این جایگاه در تعادل هاردی-وینبرگ قرار نداشت. نتایج این آزمایش نشان داد که این جمعیت چند شکلی بالایی دارد، به علاوه در بیشتر نژادهای گوسفند ایرانی مطالعه شده هر سه ژنوتیپ مشاهده نشدند ولی در این نژاد هر سه ژنوتیپ وجود داشت.

مطالعه مولکولی متیلاسیون پروموتر ژن های p14, p15 و p16 در بیماران ایرانی مبتلا به سرطان بافت سنگفرشی مری
مطالعات نشان داده است که متیلاسیون نابه جای ژنهای سرکوبگرتومور باعث خاموش شدن آنها و درنتیجه کاهش بیان آنها شده و زمینه برای ایجاد سرطان مهیا می شود. هدف از این مطالعه، بررسی متیلاسیون سه ژن سرکوبگرتومورp14,p15و p16 در مبتلایان به سرطان بافت سنگفرشی مری (ESCC) و ارتباط آن با خصوصیات دموگرافی و پاتولوژیکی بیماران می باشد. به این منظور از 44نفر بیمار ESCC غیرخویشاوند، 44عدد نمونه بافت توموری و 19عدد نمونه سالم(مجاور تومور)تهیه و مورد استخراجDNA وRNAقرار گرفت. سپس باپرایمر اختصاصی حالتهای متیله وغیرمتیله این ژنها،واکنش زنجیره ای پلیمراز ویژه متیلاسیون(MSPCR)انجام شد. بیان این ژنها نیزباRT-PCRبررسی گردید. نتایج نشان داد درمیان نمونه های توموری، درصد متیلاسیون برای ژنهایp14،p15وP16 به ترتیب عبارتست از 53%،9%و27% و در هیچ یک از نمونه های نرمال(مجاور تومور)متیلاسیونی مشاهده نشدونیزمیزان بیان این ژنها با میزان متیلاسیون نسبت عکس داشت(p<0.05). فراوانی متیلاسیون پروموتر ژنهای p15وp14در نمونه های توموری SCCE به ترتیب 41%، 9% وفراوانی وجود متیلاسیون همزمان در ژنهای p15+p16 و p14+p15 به ترتیب 6/13% و 5/4% بود درحالیکه فراوانی متیلاسیون در ژن های p14+p16، 9% مشاهده شد (P=0.0036).بدین معنا که در تمامی بیماران متیله برای ژن p14،متیلاسیون p16 مشاهده شداما در بیشتر بیمارانی کهp15متیله بود، ژن های p14ویا p16 متیلاسیون نشان نداد.مقایسه این نتایج با سایر یافته های مشابه در جهان،نشان میدهد وضعیت متیلاسیون ژن های p15،p14،p16در جمعیت ایرانی مورد بررسی در محدوده گزارشاتی از شمال چین قرار دارد. بعبارتی احتمالاً مکانیسمهای مولکولی و عوامل اتیولوژیک مشابهی میتوانند در بروز سرطان ESCC در این دو جمعیت (ایران و چین) نقش داشته باشند.

بررسی امکان رشد سلول های استخوانی(Saos-2) روی داربست های پوشش دار شده با نانوذرات کیتوزان-آلژینات
در سالهای اخیر در کشور ایران هرساله حدود 22000 مرگ و 70000 مجروح بعلت حوادث رانندگی اتفاق می افتد، که تقریبا 92٪ از مجروحین دارای شکستگی های استخوانی هستند و جهت ترمیم آسیبهای استخوانی به ارتوپدی مراجعه می کنند. طی گزارشات منتشر شده از دانشگاه علوم پزشکی شهید بهشتی، حدود 85٪ از پیوندهای مغز استخوان بعلت تهاجم باکتری ها و قارچها ناموفق بوده اند. تکنیک استفاده از داربست ها بعنوان بستر مناسبی برای تکثیر و رشد سلول ها و بافت ها از جمله راهکارهای ترمیم بافت های صدمه دیده می باشد. با این وجود استفاده از داربستهای زیست سازگار به منظور درمان نقایص استخوانی همواره با چالشهای زیادی از جمله عدم استحکام، زیست سازگاری پایین، طول عمر پایین سلولهای روی داربست بعلت تهاجم باکتری ها و قارچ ها روبرو بوده است. در این راستا از مهمترین اهداف این تحقیق، بررسی خواص سیتوتوکسیسیتی داربست های پوشش دار شده با نانوذرات کیتوزان - آلژینات، بررسی امکان رشد و حیات سلولهای استخوانی روی داربست های بدون پوشش و پوشش دار شده با نانوذرات بودند. نتایج بدست آمده نشان داد که سلول ها بر روی داربست های پوشش دار شده با نانوذرات در مقایسه با داربست های بدون پوشش بطور طبیعی رشد کردند. همچنین میزان چسبندگی سلول های استخوانی پس از 72 ساعت در این داربست ها به شکل نرمال صورت گرفت. از طرفی دیگر بررسی ساختار داربست ها با میکروسکوپ الکترونی نشان داد که داربست های پوشش دار شده با نانوذرات کیتوزان- آلژینات دارای خلل و فرج کافی برای رشد سلولهای استئوبلاست میباشند.

ارتباط پلی مورفیسم5'UTR ژن تیروگلوبولین با خصوصیات لاشه در بره های آمیخته افشاری× برولا مرینو
چربی اعماء و احشا و دنبه درصد قابل توجهی از وزن لاشه در گوسفند را به خود اختصاص می دهد. در تحقیق حاضر ارتباط پلیمورفیسم ژن تیروگلوبولین (TG) با خصوصیات لاشه در گوسفند افشاری مورد بررسی قرار گرفت. بدین منظور از تعداد 98 راس بره نر 11 ماهه نمونه خون گرفته و از آن DNA استخراج گردید. آغازگرهای لازم با استفاده از توالی ژن TG گاو طراحی شدند. محصولات حاصل از PCR با روش SSCP مورد بررسی قرار گرفت. با بررسی باندها بر روی ژل اکریل آمید، تمامی نمونهها در 6 گروه ژنوتیپی D- ,AC ,AB ,BB ,AA وEE قرار گرفتند. نتایج نشان داد که بین ضخامت چربی بین دنده 13-12 و ژنوتیپها تفاوت معنیدار )05/0>(p وجود دارد و ژنوتیپ BB کمترین (38/1cm) و ژنوتیپ D- بیشترین (87/1cm) میزان انباشت چربی در این ناحیه را دارند بالعکس درصد ضایعات به طور معنیداری در ژنوتیپ BB (20/4) از سایر گروهها بیشتر و در ژنوتیپ D- (04/3) کمتر بود. ژنوتیپ BB کمترین مقدار عمق عضله راسته (09/2)، درصد راسته (60/15)، درصد سردست (87/15) و درصد لاشه (70/42) را داشت. و همچنین ژنوتیپBB به طور معنیداری کمترین میزان درصد قلوهگاه (08/15) و وزن قبل از کشتار (40/52) را نشان داد. ژنوتیپ D- بیشترین میزان درصد قلوهگاه (94/16) وژنوتیپ AB (36/59) بیشترین میزان وزن قبل از کشتار را داشتند و اختلاف بین گروهها معنیدار بود. با توجه به این یافته می توان ژن TG را به عنوان یک ژن کاندید برای استفاده در برنامه های اصلاح نژاد این نژاد از گوسفند در نظر گرفت

امکان سنجی پیریت زدایی از زغالسنگ با استفاده از بیوفلوتاسیون
یکی از ناخالصی های همراه زغالسنگ، گوگرد است که به دو صورت آلی و معدنی وجود دارد. میزان گوگرد بیش از حد مجاز در کنسانتره زغالسنگ سبب تولید کک نامرغوب می شود. این کک در کوره های احیا مستقیم ذوب آهن، باعث تولید چدن و فولاد نامرغوب می شود. همچنین وجود گوگرد در زغالسنگ های حرارتی موجب تولید گاز دی اکسید گوگرد می شود که اثرات زیست محیطی نامطلوبی دارد. بخش عمده گوگرد معدنی زغالسنگ، پیریت می باشد. در فراوری زغالسنگ معمولا از فلوتاسیون به منظور جدایش انتخابی و بازیابی زغالسنگ از مواد باطله استفاده می شود. در روش فلوتاسیون به منظور حذف پیریت معمولا از سیانید سدیم استفاده می شود. استفاده از سیانید به دلیل سمی بودن چندان مورد توجه نمی باشد. در این تحقیق امکان پیریت زدایی از زغالسنگ طبس با استفاده از باکتری تیوباسیلوس فرواکسیدانس و روش فلوتاسیون مورد مطالعه قرار گرفت. در این روش اتصال باکتریایی، خواص سطح پیریت در زغالسنگ را از حالت آبرانی به آبدوستی تغییر می دهد. این تغییر ویژگی سطح، امکان جداسازی پیریت از زغالسنگ را در فلوتاسیون میسر می سازد. باکتری ها در محیط کشت 9K، کشت داده شد. آزمایش های بیوفلوتاسیون در سلول فلوتاسیون Denver انجام شد و تاثیر عوامل مختلف از قبیل درصد جامد پالپ، اندازه ذرات زغالسنگ، دانسیته باکتریایی و مدت زمان ماند بر حذف پیریت مورد بررسی قرار گرفت. نتایج نشان داد که با استفاده از این روش، پیریت به میزان متوسط 62 درصد و گوگرد کلی به میزان متوسط 35 درصد کاهش می یابد.

شناسایی مکان های ژنی کنترل کننده زمان گلدهی در توتون تیپ شرقی
به منظور شناسایی مکان های ژنی مرتبط با گلدهی در توتون تیپ شرقی، جمعیت ژنتیکی شامل ۱۰۰ فرد 2F حاصل از تلاقی دو ژنوتیپ توتون شرقیSPT 406 (والد پدری) وBasma Seres 31 (والد مادری) برای صفت روز تا شروع گلدهی مورد ارزیابی قرار گرفتند. در آزمایشات مولکولی نقشه پیوستگی جمعیت 2F با ۲۳ نشانگر SSR و ۲۹ نشانگرISSR تهیه گردید که cM 570.8 از ژنوم توتون را پوشش می داد. با استفاده از روش های مکان یابی فاصله ای ساده و مرکب به ترتیب ۹ و ۲ QTL برای صفت مورد مطالعه شناسایی گردید. در این مطالعه، بیشترین تعداد QTL در گروه پیوستگی شماره پنج شناسایی شد. QTL های qDF5-3 و qDF2-2، شناسایی شده با روش مکان یابی فاصله ای ساده، به ترتیب با توجیه 0.8 و ۳۶ درصد از تغییرات فنوتیپی صفت )2(R، به ترتیب کوچکترین و بزرگترین QTL های شناسایی شده می باشند. درصد تغییرات فنوتیپی توجیه شده )2(R توسط QTL های شناسایی شده با روش مکان یابی فاصله ای مرکب (qDF5-1 و qDF5-2)، به ترتیب 1.95 و 15.34 درصد بود. نتایج نشان داد که مکان هایی با اثرات ژنی افزایشی و غالبیت در کنترل ژنتیکی صفت روز تا شروع گلدهی در توتون تیپ شرقی نقش دارند.

بیان هترولوگ آنزیم کیتیناز 36 کیلو دالتونی از قارچ Trichoderma atroviride در میزبان پروکاریوتی
آنزیمهای کیتینولیتیک در تجزیه پسماندهای کیتینی نظیر پوسته سختپوستان و دیواره سلولی قارچها و تولید الیگوساکاریدها از کیتین نقش دارند. خصوصیات کاتالیتیک آنها، این آنزیمها را هدف مناسبی جهت استفاده درصنعت یا به عنوان بیوکنترل قرار داده است. در این مطالعه جهت تکثیر و همسانهسازی ژن chit36 از جدایه بومی ایران Trichoderma atroviride PTCC5220، آغازگرهای اختصاصی (chit36pf/chi36r) طراحی و پس از تکثیر قطعه مورد نظر، به منظور تولید پریپلاسمی در وکتور pET26b(+) همسانهسازی و جهت بیان به باکتری E. coli BL21-DE3 انتقال داده شد. در بررسی نتایج حاصل از SDS-PAGE هیچگونه باندی دال بر بیان پروتئین در هیچکدام از شرایط دمایی و میزان متفاوت IPTG در بخشهای متفاوت سلولی مشاهده نشد. لذا در ادامه، بیان پروتئین Chit36 با حذف پپتید نشانه pelB (جهت تولید به صورت سیتوپلاسمی) همراه با توالی هیستیدین در انتهای کربوکسیلی دردستور کار قرارگرفت. باکتری حاوی سازه نوترکیب تحت شرایط القایی متفاوت مورد بررسی قرار گرفت. علیرغم عدم مشاهده هیچگونه باندی در هیچکدام از شرایط القایی با روش SDS-PAGE، بیان اندک پروتئین در 2 کلنی با استفاده از روش Western blot مورد تایید قرار گرفت. به منظور بهینه سازی شرایط بیان، میزان بیان پروتئین نوترکیب نسبت به پروتئین تام باکتری درشرایط متفاوت القایی، به روش سنجش کمی باند های پروتئینی روی ژل SDS-PAGE مورد ارزیابی قرار گرفت. نتایج حاصله نشان داد که بهترین شرایط بیان در زمان القا OD600 : 0.3 و میزان IPTG یک میلیمولار و زمان انکوباسیون 6 ساعت میباشد، دراین شرایط میزان بیان پروتئین نوترکیب نسبت به پروتئین تام برابر %57/ 45 میباشد.
