خالص سازی وتعیین خصوصیات کینتیکی آنزیم آسپاراژیناز در باکتری Serratia marcescens
آسپاراژیناز یک آنزیم هیدرولاز می باشد که آسپاراژین را به آمونیوم و آسپارتیت تبدیل می کند. این آنزیم کاربرد مؤثری در درمان لوسمی لنفوبلاستیک دارد. marcescens Serratia یک باکتری گرم منفی است که کلنیهای آن دارای رنگیزه قرمز می باشند که می تواند در صورت حضور آسپاراژین در محیط کشت خود، تولید آنزیم آسپاراژیناز نماید. در این تحقیق آنزیم آسپاراژیناز از محیط کشت Serratia خالص سازی و همچنین فاکتور های کینتیکی آن تعیین شد. باکتری Serratia در محیط مایع حاوی پپتون، عصاره مالت و آسپاراژین کشت شد و محیط بعد از 44 ساعت سانتریفیوژ شد و محلول رویی که دارای آنزیم آسپاراژیناز بود برای مطالعات کینتیکی و خالص سازی مورد استفاده قرار گرفت. مراحل تخلیص آنزیم با استفاده از رسوب دهی با آمونیوم سولفات و به دنبال آن دیالیز و ستون کروماتوگرافی DEAE سلولز انجام شد. انجام الکتروفورز به روش SDS-PAGE باندی با وزن مولکولی تقریبی حدودkDa 54 را بر روی ژل نشان داد که نشان دهنده خلوص آنزیم بود. مقایسه تأثیر تغییرات pH (10-4) بر فعالیت آنزیم نشان داد که بیشترین فعالیت آنزیم در 7 pH است، و بررسی تغییرات دما بر فعالیت آنزیم نشان داد که بیشترین فعالیت در دمای 40 درجه سانتی گراد می باشد. همچنین مقدار فاکتور های کینتیکی Vmax و Km در این آنزیم مشخص گردید.

مخمر پیکیا پاستوریس: ابزار آزمایشگاهی مناسب برای تولید پروتئین های نوترکیب
سیستم بیانی باکتری توانایی تولید مقادیر بالایی از پروتئین های نوترکیب را داراست، اما ممکن است پروتئین های تولید شده تاشدگی ساختاری مناسبی نداشته و فاقد اصلاحات پس از ترجمه ای باشند. از طرف دیگر سلول های پستانداران اگرچه انتخاب مناسبی برای تولید پروتئین نوترکیب محسوب می شوند اما هزینه های بالای تولید و میزان کم محصول نوترکیب در این سیستم ها، محققان را به سمت مطالعه بر روی سیستم بیانی مخمری سوق داده است. مخمرها به دلیل داشتن ویژگی های بیشمار ازجمله دستکاری ژنتیکی آسان، میزان بالای بیان پروتئین های برون سلولی و درون سلولی، و توانایی انجام اصلاحات پس از ترجمه ای متناسب با سلول های یوکاریوتی، برای بیان پروتئین های خارجی مناسب هستند. مخمر متانول دوست پیکیا پاستوریس برای تولید پروتئین های نوترکیب در تراکم سلولی بالا و محیط کشت ارزان، توسعه یافته است. ترشح مقادیر کمی از پروتئین های درونی مخمر در محیط کشت، باعث خالص سازی آسان پروتئین های نوترکیب از آن می شود. به هرحال، سویه های گلایکوسویچ مخمر پیکیا پاستوریس با قابلیت انجام اصلاحات پس ترجمه ای یکسان با سیستم بیانی یوکاریوتی، تنها نقص سیستم های بیانی مخمر را رفع کرده است. بر این اساس، پیکیا پاستوریس نسبت به سایر میزبان ها، سیستم ترشحی مناسبی برای دستیابی به مقادیر بالاتری از پروتئین های نوترکیب با ساختار درست بشمار می رود.

طراحی ترکیبات کرومنی جدید با فعالیت ضد سرطانی و بررسی چگونگی میانکنش آنها با توبولین به روش داکینگ مولکولی
در حال حاضر روشهای in silico یکی از کم هزینهترین و سریعترین روشهای موجود جهت طراحی و کشف دارو در زمینه درمان محسوب می گردد. در این مطالعه ما علاقهمند شدیم تا با انجام طراحی محاسباتی دارو به روش داکینگ مولکولی ترکیباتی را معرفی کنیم که نقش موثری در مهار پلیمریزاسیون توبولینها به عنوان عوامل اصلی تقسیم سلولی در تومورهای سرطانی دارند. بدین منظور ترکیبات کرومنی که توسط محققین مختلف خواص ضد سرطانی آنها مورد آزمایش قرار گرفته بودند، جمع آوری، و بر اساس جایگاه اتصال آنها به توبولین، آنالوگهای جدیدی طراحی شدند. سپس، با استفاده از روش داکینگ مولکولی توسط نرم افزار آکادمیک AutoDock Vina ترکیبات قویتر شناسایی شدند و برهمکنشهای احتمالی این ترکیبات با جایگاه اتصال کلشیسین در توبولین آنالیز شد. با توجه به اینکه ترکیبات طراحی شده با مقدار انرژی پایینتری نسبت به کلشیسین به ساختار پروتئین داک شدند، میتوانند به عنوان ترکیبات بالقوه دارویی مورد ارزیابیهای بعدی قرار بگیرند.

تنوع ژنتیکی توس (Betula pendula)، که یک گونه ارزشمند دارویی و در معرض خطر انقراض است، در سطح اندک جمعیت های باقیمانده از آن در نیمرخ شمالی ایران شامل: مارمیشو، سیاه مرزکوه، سنگده و شهرستانک، با استفاده از پلی مورفیسم DNAی کلروپلاستی سه منطقه 3800، 1800 و 2700 جفت بازی و تکنیک PCR-RFLP مطالعه شد. این مناطق به ترتیب CD ، DT و K1K2 نامیده می شوند. درصد جایگاه پلی مورفیک، هتروزیگوتی مورد انتظار (He) و شاخص شانون (I) در چهار جمعیت، به ترتیب برابر با 51/24 درصد، 096/0و 14/0 محاسبه شد. آنالیز واریانس مولکولی نیز نشان داد که 66 درصد از کل تنوع مربوط به درون جمعیت ها و 34 درصد آن بین جمعیتی است. محاسبه شاخص تمایز ژنتیکی (206/0Gst =)، تمایز بالای جمعیتهای توس از یکدیگر را نشان داد. مقایسه دو به دو میزان تمایز ژنتیکی جمعیتها از یکدیگر نشان داد که جمعیت مارمیشو بیشترین تمایز را از سایر جمعیت ها نشان داد. برآورد میزان جریان ژنی بین جمعیتها حاکی از وقوع رانش ژنتیکی بین آنها است (96/0= (Nm. همچنین نتایج آزمون مانتل همبستگی معنیداری (77%r= ) را بین تمایز ژنتیکی جمعیت و فاصله جغرافیایی آنها از یکدیگر نشان داد. نتایج تحقیق حاضر وقوع رانش ژنتیکی در جمعیت های توس ایران و خطر در معرض انقراض بودن این گونه را تایید می کند. که بهکارگیری راهکاری سریع و مناسب برای حفاظت آن را تاکید می نماید.

کلونینگ، بیان و تعیین خصوصیات لیپاز کایمریک باسیلوس ترموکاتنولاتوس در باکتری Escherichia coli
لیپازهای باکتریایی عضوی از خانواده β/α هیدرولازها هستند که تری آسیل گلیسرول ها را در فاز بین آب-چربی هیدرولیز می کنند. لیپاز باکتری باسیلوس ترموکاتنولاتوس (BTL2) در دمای 75-60 درجه سانتیگراد و pH برابر 10 -8 فعالیت دارد و مناسب استفاده در صنعت شوینده می باشد. در این پژوهش لیپاز باسیلوس ترموکاتنولاتوس کایمریک حاوی توالی توافق شده لیپاز قارچ کاندیداروگوزا (207Gly-Glu-Ser-Ala-Gly211)در ناحیه زانوی هسته دوست، در باکتری E. coli کلون و بیان شد. سپس فعالیت آنزیمی لیپاز کایمریک در حضور سوبستراهای مختلف اندازهگیری شد. همچنین اثر عوامل مختلف از قبیل دما، pH، دترجنتها، حلالهای آلی و یونهای فلزی بر روی فعالیت آنزیم بررسی گردید .نتایج نشان داد که آنزیم کایمریک بیشترین فعالیت را در حضور سوبسترای 4 کربنه، (pH 9.0) و دمای 60 درجه سانتیگراد دارد. همچنین فعالیت آنزیم در حضور حلال های آلی N- هگزان، N- هپتان، متانول و کلروفرم و دترجنت های ترایتون 100-X ، توین 20، توین 40 افزایش یافته است در حالیکه یون های فلزی عمدتاً اثر کاهشی بر فعالیت آنزیم کایمریک داشتند.

مطالعه توالی، خاصیت آنتیژنی و ساختار سه بعدی پروتئین های غیراختصاصی ناقل لیپید2 در برنج ایرانی: ارزیابی بیوانفورماتیکی
پروتئینهای غیراختصاصی ناقل لیپید 2(nsLTP2) یکی از اعضای خانواده پرولامینها هستند که به دلیل ساختار ویژه خود توانایی انتقال ترکیبات لیپوفیل مختلفی را دارند. با وجود مزیتهای نسبی پروتئین LTP2 در انتقال دارو، خاصیت آلرژنی این پروتئینها نیز گزارش شده است. هدف مطالعه حاضر بررسی توالی و ساختار پروتئین nsLTP2 برنج ایرانی و نیز بهبود برخی از ویژگی های آن برای استفاده در سیستمهای تحویل دارو، توسط ابزارهای بیوانفورماتیک است. مقایسه توالی پروتئین های nsLTP2 انواع برنج، حداقل 51% شباهت بین گونههای مختلف را نشان میدهد. بر اساس ارزیابی های فیلوژنی مشخص شد که بیشترین شباهت بین توالی ژن این پروتئین با گونه ژاپنی (Accession NO. NP 0011048723.1) وجود دارد. موتیفها و جایگاههای مختلف nsLTP2 برنج ایرانی با کمک آنالیز توالی به دست آمد. همچنین مطالعات بیوانفورماتیکی نشان دهنده 4 جایگاه با خاصیت شبهآنتیژنی است که 3 عدد آن بعد از تغییرات پس از ترجمه باقی میماند و حضور آنها میتواند خاصیت آلرژنی این پروتئینها را توجیه کند. همچنین، بررسی توالی دو گونه ژاپنی و ایرانی موید این است که اسیدهای آمینه شرکت کننده در جایگاه فعال، به-طورکامل حفاظت شده است. بهمنظور مطالعه تاثیر حذف توالیهای آلرژن بر تمایل به لیگاندهای هیدروفوب، از روش مدلسازی در راستای تعیین ساختار، استفاده گردید. نتایج تحقیق حاضر نشان داد که در صورت اعمال تغییراتی در توالی و حذف بخشهای آلرژن nsLTP2، احتمالا میتوان بازده پروتئین را در سیستمهای تحویل دارو افزایش داد.

تنوع ژنتیکی جمعیت نزد گونه های جنس Rattus در شهر تهران
رت ها امروزه از یک طرف مدل آزمایشگاهی بسیار مهمی به حساب می آیند و از طرف دیگر یک آفت بسیار مهم و بالقوه خطرناک می توانند باشند. مطالعه در مورد رت ها بخصوص برای شناسایی آنها، ژنتیک جمعیت شان و جغرافیای تکاملی آنها با استفاده از روش های مورفولوژیک و بالاخص مولکولی یک ضرورت است. این پژوهش با هدف شناسایی و بررسی ژنتیک رت های کلان شهر تهران با استفاده از ژنوم میتوکندری در ناحیه D-Loop انجام گرفت و نتایج آن نشان داد که در شهر تهران فقط گونه های رت سیاه و قهوه ای وجود دارد. از میان 229 نمونه فقط یک نمونه رت سیاه و در بین رت های قهوه ای نیز دو زیرگونه احتمالی کاملا متفاوت تشخیص داده شد. نتیجه آنالیزهای ژنتیک جمعیت و دموگرافیک نشان داد که این جانوران دارای تنوع ژنتیکی خیلی پایینی هستند و دو گروه متفاوت با زمان مختلفی وارد ایران شده اند: ورود گروه اول که جدیدتر است حدود 700 و گروه قدیمی 5600 سال پیش تخمین زده شد. داده ها همچنین نشان داد که جمعیت رت های تهران در حال رشد و افزایش حجم است ولی در گذشته کنترلهای خوبی انجام شده است. مناطقی از تهران مثل 18، 19،13، 12، 9، 4 و 1 دارای جمعیت با تنوع ژنتیکی خیلی بیشتری از رت ها هستند و میتوانند محل اصلی کلنی ها باشند.

بهینه سازی منبع ازت و میزان اکسیژن محلول برای تولید همزمان اتانول و زایلیتول در کشت همزمان دو مخمر Saccharomyces cerevisiae و Candida tropicalis
یکی از راهکارهای جدید و امید بخش برای اقتصادی سازی تولید بیواتانول از پسماندهای لیگنوسلولزی ، تولید همزمان یک یا چند ماده با ارزش دیگر در کنار اتانول از پسماند مد نظر (تصفیه زیستی) می باشد. هدف از اجرای تحقیق حاضر بهینه سازی تولید همزمان اتانول و زایلیتول در کشت همزمان دو مخمر Saccharomyces cerevisiae و Candida tropicalis در محیط کشت طراحی شده در سطح فرمانتور غیر پیوسته بود. بمنظور تعیین بهترین منبع ازت برای دو سویه، ابتدا دو سویه در 8 منبع ازت آلی و معدنی مختلف (6 گرم در لیتر) در قالب طرح یک فاکتور در یک زمان بصورت همزمان کشت داده شدند. نتایج بدست آمده نشانگر اختلاف معنی دار منابع مختلف از نظر کارایی بود و عصاره مخمر بالاترین میزان اتانول (41/24 گرم در لیتر)، زایلیتول (7/22 گرم در لیتر) و زیست توده (24/13 گرم در لیتر) را نشان داد. در ادامه تاثیر غلظت های مختلف اکسیژن محلول شامل 5%، 10%، 20% و 30% بر کارایی تولید دو محصول در سطح فرمانتور غیر پیوسته مورد بررسی قرار گرفت. بیشترین میزان اتانول تولیدی(38 گرم در لیتر) با با بازدهی 47/0 (گرم اتانول تولیدی بر گرم گلوکز مصرف شده) در اکسیژن محلول 5% مشاهده شد، در حالی که بیشترین میزان زایلیتول تولیدی به میزان 6/21 گرم در لیتر با بازدهی 54/0 (گرم زایلیتول تولیدی نسبت به گرم زایلوز مصرف شده) در اکسیژن محلول 10% به دست آمد. همچنین بیشترین میزان بیومس تولیدی با میزان 2/25 در بالاترین میزان اکسیژن مورد آزمایش یعنی 30% بدست آمد.

شرایط استخراج روی توان آنتی اکسیدانی و ترکیب بیوشیمیایی آرتیمیزیا آفسنطین
اثرات دما و نوع حلال بر روی استخراج ترکیبات بیوشیمیایی و ظرفیت آنتی اکسیدانی Artemisia absinthium مطالعه شدند. تغییر مقدار ترکیبات بیوشیمیایی و ظرفیت آنتی اکسیدانی عصاره¬ها در شرایط¬ استخراجی متفاوت شامل دما (60-30 درجه سانتیگراد)، حلال (متانول اتانول و استونیتریل) و غلظت¬ حلال (100-25 %) مطالعه شد. مقادیر کل فنل¬ (mg GAE/100 g DW 1033-612) و توتال فلاونوئید (mg CE/100 g DW 392) در نمونه¬ها اندازه گیری شد. ظرفیت شکار رادیکال های آزاد با استفاده از روش های مختلف، DPPH (2,2-diphenyl-1-picrylhydrazyl) (79-51%)، FRAP (ferric reducing antioxidant power) (µM TE /100g DW287-151) و ABTS (2,2-azinobis 3-ethyl-benzothiazoline-6-sulfonic acid) (39-17 mM TE/g FM) اندازه گیری گردید. بر اساس نتایج بدست آمده، متانول به عنوان حلال مناسب استخراج انتخاب شد. عصارهای بدست آمده در شرایط متانول 75 درصد و دمای 45 درجه بالاترین فعالیت آنتی اکسیدانی را نشان دادند، در حالی که عصارهای بدست آمده با استفاده از متانول 75 درصد و دمای 60 درجه بالاترین سطح فنل را داشتند. آنالیز نمونه¬ها برای ترکیب آرتمیزینین با استفاده از روش کروماتوگرافی مایع با کارایی بالا (HPLC) انجام شد اما برخلاف برخی مطالعات، این ترکیب در نمونه¬ها شناسایی نشد . نتایج HPLC وجود بیشترین مقدار ترکیب anabsinthin (58/16 میکروگرم در گرم گیاه) را در متانول 75% و کمترین مقدار ( 45/9 میکروگرم در گرم گیاه ) را درمتانول 25% نشان داد.

همسانه سازی و بررسی بیان ژن متالوتیونین smtA در باکتری اشریشیا کلی به منظور جذب فلزات سنگین
یکی از راه های جذب فلزات سنگین علاوه بر روش های شیمیایی و فیزیکی، استفاده از فرآوردههای زیستی می باشد. یکی از این فرآورده ها، پروتئین های جاذب فلزات سنگین مانند متالوتیونین می باشد.هدف: هدف از این مطالعه، همسانهسازی ژن متالوتیونین smtA از سیانوباکتری سینکوکوس ایلانگاتوس سویه PCC7942 ، بیان و بررسی جذب فلز توسط آن میباشد.روشها: ژن smtA در ناقل همسانهسازیT/A ، همسانهسازی و در باکتری E.coli (DH5α)تراریخت شد. صحت تراریختی با Colony-PCR بررسی شد. سپس ژن مورد نظر به داخل ناقل بیانی pET15b زیرهمسانه سازی و در باکتری E.coli (BL21) تراریخت گردید. صحت تراریختی با بررسی الگوی هضم آنزیمی و تعیین توالی تایید گردید. القای بیان ژن توسط IPTG انجام شد و بیان پروتئین توسط تریس-تریسین SDS-PAGEو وسترن بلات بررسی شد. بررسی جذب یون کادمیم توسط پروتئین توسط دستگاه جذب اتمی صورت گرفت.نتایج: نتایج توالییابی، صحت تراریختی را تائید کرد. بیان پروتئین تولید شده با روش های SDS-PAGEو وسترن بلات تایید گردید. بررسی جذب فلز نشان داد که یک میلی گرم پروتئین SmtA می تواند به میزان 28 نانو مول یون کادمیم را جذب نماید.

بررسی نحوه تاثیر جهش ها بر غیر فعال شدن آنزیم پیرازین آمیداز با شبیه سازی دینامیک مولکولی
پیرازین آمید یکی از مهم ترین داروها برای کنترل مایکوباکتریوم توبرکلوزیس، عامل بیماری سل، است. ژن pncA آنزیم پیرازین آمیداز مایکو باکتریوم توبرکلوزیس را رمزدهی می نماید. این آنزیم مسئول تبدیل داروی پیرازین آمید به شکل فعالش یعنی پیرازینوئیک اسید است. علیرغم نقش این دارو در کوتاه سازی دوره ی درمان از نه ماه به شش ماه، ظهور سویه های مقاوم به پیرازین آمید مشکل مهم سلامت جهانی است. در این مطالعه دو ژن پیرازین آمیدازجهش یافته(D63G/W119Cو T160P)از سویه های مقاوم پیرازین آمیدی و نیز پیرازین آمیداز نوع وحشی از سویه ی مرجع H37Rvهمسانه سازی، بیان، تعیین توالی شده و تعیین فعالیت شدند. با استفاده از همولوژی مدل سازی و جایگزینی آمینواسیدی، ساختار پیرازین آمیدازهامورد مطالعه قرار گرفت. نتایج نشان داد که دوجهش یافته یT160P وD63G/W119Cفعالیت پیرازین آمیدازی خود را به طور کامل از دست داده اند. نتایج محاسباتی نیز تأیید کرد که فقدان فعالیت این آنزیم ها عمدتا به دلیل تاثیر موضعی جهش های ایجاد شده در ساختار دوم (در اکثر موارد ساختار های آلفا هلیکس) اطراف محل جهش ها است که موجب تغییر ساختاری شده است. این تغییراحتمالا موجب تغییر ساختار سه بعدی جایگاه فعال در مورد جهش یافته D63G/W119C وکاهش ابعاد دهانه ی پاکت اتصال در جهش یافته T160P شده است.

بهینه سازی شرایط بیان آنزیم کیتیناز42 کایمری در میزبان پروکاریوتی و مقایسه فعالیت آن با آنزیم کیتیناز42
آنزیم¬های کایمری از جمله پروتئین¬های مهندسی شده می¬باشند که از تغییر در ساختار آنزیم¬ها بدست می¬آیند. آنزیم¬های کیتینازی بدلیل توانایی در تجزیه ترکیبات کیتینی، در تولید پروتوپلاست قارچی، تبدیل زیستی shellfish waste به بخشهای تک جزئی، تولید الیگوساکاریدها و کنترل قارچ¬های بیماریزا حائز اهمیت می-باشند. در این تحقیق پس از تکثیر ناحیه (chitin binding domain) ChBD کیتینازB باکتری Serratia marcescens و cDNA ژن کیتیناز42 قارچ Trichoderma atroviride (فاقد ChBD) با استفاده از آغازگرهای همپوشان، کیتینازکایمری با استفاده از روش SOEing PCR تکثیر و در ناقل بیانی همسانه¬سازی شد. سازه بیانی نوترکیب حاصل ابتدا به میزبان بیانی پروکاریوتی منتقل و سپس بهینه سازی بیان پروتئین مورد نظر در این میزبان، با استفاده از روش تاگوچی انجام شد. اثر عواملی مانند غلظت IPTG، دمای انکوباسیون و مدت زمان انکوباسیون پس از القاء، بر بیان کیتینازکایمری بررسی شد. با توجه به آرایه¬های متعامد تاگوچی در 3 عامل و 4 سطح، 16 آزمایش طراحی شد. پس از انجام این آزمایش ها پروتئین تام از سلول استخراج و در ژل SDS-PAGE مورد بررسی قرار گرفت. نتایج حاصل از SDS-PAGE با استفاده از نرم افزار Total lab کمی شده و سپس با استفاده از برنامه Qualitek-4 تجزیه وتحلیل شد. نتایج حاصل نشان داد که در بین عوامل مختلف، غلظت IPTG بیشترین اثر را بر بیان پروتئین کیتینازکایمری دارد. بهترین شرایط بیان برای پروتئین کیتینازکایمری به ترتیب IPTG با غلظت یک میلی¬مولار، دمای 28 درجه سانتیگراد به مدت 16 ساعت بدست آمد.

قارچ ها به عنوان یکی از عوامل فرسودگی زیستی بنای آرامگاه کوروش کبیر
پاسارگاد، پایتخت سلسله¬ی هخامنشیان در زمان کوروش کبیر است که زمان ساخت آن به قرن ششم قبل از میلاد مسیح برمی¬گردد. از مهم ترین آثار باستانی پاسارگاد، مقبره ی کوروش کبیر است که عمده آن از سنگ¬های آهکی ساخته شده است. پاسارگاد، دومین بنای تاریخی بزرگ بعد از پرسپولیس می باشد. این بنا توسط UNESCO به عنوان میراث جهانی شناخته شده است. این مطالعه به شناسایی قارچ های عامل فرسودگی زیستی این بنا که برای اولین بار در ایران انجام گرفته است می پردازد. بطور کلی با استفاده از محیط های کشت Potato Dextrose Agar و Sabouraud Dextrose Agar، 33 جدایه از سطوح سنگی آرامگاه به¬دست آمد. از جدایه ها کشت خالص تهیه شد. برای مشاهده میکروسکوپی ساختارهای زایشی و رویشی قارچ، از تکنیک اسلاید کالچر استفاده شد و قارچ ها بر اساس خصوصیات شکلی میکروسکوپی و ماکروسکوپی شناسایی شدند. در این بررسی، قارچ¬های رنگی و مخمر ها جدا شدند. قارچ هایAlternaria sp. Cladosporium sp., Ulocladium sp., Fusarium sp., Humicula sp., Arthirinium sp. و مخمر ها، قارچ های غالب جدا شده از سطوح مذکور بودند. میکروسکوپ الکترونی نگاره (SEM) برای مشاهده ی روند فرسودگی زیستی ناشی از رشد قارچ استفاده شد و پدیده هایی مانند ایجاد حفره های زیستی، شکرزدگی و سنگ شویی به خوبی قابل رویت بود. این پدیده ها در اثر رشد میسلیوم قارچ ها و نفوذ آن به درون سنگ و ترشح متابولیسم¬های قارچی رخ می دهند. جداسازی و شناسایی جدایه ها، اولین قدم در محافظت از سطوح آثار باستانی می¬باشد.

اثرات عصاره آبی و اتانولی گل محمدی (Rosa damascena mill L.) بر علیه سلولهای سرطانی معده انسان
مقدمه: سرطان معده یکی از عوامل مهم بیماری و مرگ و میر در جهان است. گونههای گیاهی منحصر به فرد زیادی وجود دارد که برای یافتن ترکیبات ضد سرطانی باید مطالعه شوند. هدف: در این تحقیق، خاصیت ضد سرطانی عصاره اتانولی و آبی گل محمدی بر رده سلولهای سرطانی معده انسان (AGS) بررسی شد. روش بررسی: هشت غلظت متفاوت از عصارهها به همراه داروی 5- فلورواوراسیل بر روی رده سلولی سرطان معده اعمال شد. سمّیت عصارهها با آزمون MTT و اثر عصارهها بر تکثیر سلولی با سنجش مصرف BrdU بررسی گردید. روش TUNEL برای اندازهگیری مرگ آپوپتوزی سلول بهکار رفت.نتایج: نتیجه نشان داد عصاره آبی و عصاره اتانولی گل محمدی بقای سلولهای سرطانی را بهطور معنیداری کاهش میدهند. شاخص IC50 برای این دو عصاره روی سلول AGS به ترتیب 887/3 و 517/2 میکروگرم بر میلیلیتر به دست آمد. نتایج روش میزان مصرف BrdU نشان داد هر دو عصاره آبی و اتانولی سبب کاهش معنیدار تکثیر سلولهای سرطان معده (نسبت به سلول فیبروبلاست) میشوند. با افزایش غلظت هر دو عصاره میزان تکثیر نیز کاهش مییابد. همچنین اثر کشندگی و مهاری عصاره اتانولی بیشتر از عصاره آبی است. میزان بروز مرگ آپوپتوزی ناشی از افزودن هر دو عصاره آبی و اتانولی روی سلولهای سرطانی 90% برآورد گردید.نتیجه گیری: عصاره آبی و اتانولی گل محمدی از طرق مختلف باعث کاهش بقا و تکثیر و القای مرگ در سلول سرطانی معده انسان میشود.

انتقال T-DNA وایجاد گیاه تراریخت داتوره (Datura metel L.)
در این تحقیق، شرایط ایجاد گیاه تراریخت از Datura metel L با استفاده از A. tumefaciens حامل پلاسمید pZM1047 شامل ژنهای NPTII، و بتا گلوکورونیداز (GUS) بهینه گردید. از سوسپانسیون باکتری جهت انجام co-culture با قطعات برگ استفاده شد و سپس قطعات گیاه به محیط باززایی MS حاوی ١ میلی گرم در لیتر هورمون BAP، 700 میلی گرم در لیتر سفازولین ، 25 میلی گرم در لیتر کانامایسین منتقل گردیدند. بعد از٢٠-١٥ روز کالوسهای در حال باززایی ظاهر شدند و بعد از انتقال ساقه باززایی شده به محیط MS بدون هورمون ریشه دار شدند. انتقال ژن در گیاهان تراریخت با استفاده از PCR مورد تأیید قرار گرفت. نرخ ترانسفورماسیون حدود 13 درصد بهدست آمد. که نسبت به درصد گزارش شده توسط محققان قبلی 2 درصد افزایش داشت.در این تحقیق، شرایط ایجاد گیاه تراریخت از Datura metel L با استفاده از A. tumefaciens حامل پلاسمید pZM1047 شامل ژنهای NPTII، و بتا گلوکورونیداز (GUS) بهینه گردید. از سوسپانسیون باکتری جهت انجام co-culture با قطعات برگ استفاده شد
