معرفی سویه جدید مخمری Candida sp. strain MY2 با توان بالقوۀ سنتز سریع و برون سلولی نانوکریستال های اکسید روی
میکروارگانیسم های محتلف به عنوان نانوکارخانه های سبز به علت توان بالقوۀ فوق العاده خود قادر به ایجاد محیط سبز به منظور ایجاد نانوساختارهای کنترل شده هستند. نانوذرات اکسید روی با توجه به توان بالقوۀ بالا در انجام فرآیندهای درمانی اختصاصی، در مطالعات علوم زیستی، دارویی و پزشکی کاربرد فراوان دارند. در این پژوهش، غربالگری سویه های مخمری با امکان استفاده از آنها به عنوان بیوکاتالیزور در تولید برون سلولی نانوذرات اکسید روی مورد بررسی قرار گرفت. از میان 7 سویه مخمری غربالگری شده از خاک های معادن مس، سویه MY2 دارای توان بالقوۀ بالا در سنتز برون سلولی نانوذرات اکسید روی بود. سویه مذکور از نظر صفات فنوتیپی و ملکولی شناسایی و در جنسCandida (با شماره دسترسیKF560329) رده بندی شده است. تشکیل اولیه نانوذرات توسط آنالیز نوری UV-visible بررسی شد. الگوی پراش اشعه ایکس نانوذرات حاصل نشان داد که ذرات سنتز شده به صورت نانوکریستال های اکسید روی هستند. مورفولوژی و توزیع اندازه نانوذرات به وسیله میکروسکوپ الکترونی روبشی (Scanning electron microscopy) و هیستوگرام پراکنش اندازه نانوذرات مشخص گردید. یافته های به دست آمده توان بالقوۀ بالای سویه مخمری Candida sp. strain MY2 در تولید سریع و برون سلولی نانوذرات اکسید روی را نشان می دهد به طوری که می توان از ترشحات این مخمر به عنوان بیوکاتالیزور ایمن و ارزان قیمت در تولید نانوکریستال های اکسید روی با توزیع نسبتا باریک اندازۀ ذرات (5-50 نانومتر) و متوسط اندازۀ ذرات 5/23 نانومتر پس از 8 ساعت واکنش زیست تبدیلی بهره جست.

بررسی میزان مقاومت کلی فرم های روده ای جدا شده از پساب های صنعتی، خانگی و بخش هایی از تصفیه خانه شهر اراک به فلزات سنگین
فلزات سنگین نظیر کادمیوم، جیوه، نیکل و اکسی آنیون های تلوریت و کرومات از عناصر سمی هستند که به طرق مختلف وارد محیط زیست شده و برای سیستم¬های بیولوژیکی، بالاخص انسان، حیوانات و گیاهان زیان آور می باشند، لذا حذف آن ها از محیط زیست ضروری است. میکروارگانیسم ها و به ویژه باکتری ها نقش بسیار مهمی در حذف زیستی فلزات سنگین داشته و در بیوتکنولوژی کاربرد دارند. در این پژوهش شمارش کلی فرم های روده ای و جداسازی سویه های مقاوم به فلزات سنگین از پساب های صنعتی، خانگی و قسمت های مختلف سیستم تصفیه پساب شهر اراک انجام شده است. در این مطالعه چهار جنس از خانواده باکتری های روده ای شامل Klebsiella spp، Escherichia coli، spp Citrobacter و spp Enterobacter از نمونه پساب های فوق جداسازی شدند و حداقل تراکم متوقف کننده رشد MIC)در مقابل غلظت های مختلف فلزات سنگین نظیر کلرید کادمیوم، سولفات نیکل، کلرید جیوه، کرومات پتاسیم و تلوریت پتاسیم با استفاده از روش رقت در آگار، تعیین گردید و مقاومت آنها در برابر تعدادی از آنتی بیوتیک ها با روش دیسک متد مشخص شد. در این تحقیق برای اولین بار MIC بسیار بالا ی 22mM نسبت به کادمیوم، 20mM نسبت به کروم، 20mM نسبت به تلوریت، 10mM نسبت به نیکل و 1mM نسبت به جیوه برای باکتری Klebsiella spp بدست آمد.

بررسی تحمل و جذب فلزات مس و سرب توسط سه سویه استاندارد مخمر
آلودگی محیط زیست توسط فلزات سنگین به یکی از جدیترین معضلات زیست محیطی امروز تبدیل شده است. در طی سالیان گذشته روشهای مختلف فیزیکی و شیمیایی برای پاکسازی فلزات سنگین مورد بررسی قرار گرفتهاند، اما اکثر آنها مشکل، گران و انرژی بر هستند. استفاده از میکروارگانیزمها برای جذب فلزات سنگین، میتواند جایگزینی مناسب برای این روشها باشد. در این تحقیق از سه مخمر استاندارد PTCC 5052 Saccharomyces cerevisiae، Kluyveromyces marxianus DSMZ 5422،Yarrowia lipolytica DSMZ 8218، برای بررسی مقاومت نسبت به غلظتهای مختلف فلزات سنگین مس و سرب استفاده شد. مقاومت بیشتری نسبت به مس (یاروویا لیپولیتیکا تا 6 میلیمول، کلویورومایسز مارکسیانوس تا 5 میلیمول و ساکارومایسز سرویزیه تا 1 میلیمول) و مقاومت کمتری نسبت به سرب (هر سه مخمر تا غلظت 2 میلیمول) مشاهده شد. سپس بر اساس مقاومتهای مشاهده شده توانایی مخمرها در جذب این فلزات سنگین مورد آزمایش قرار گرفت. میزان جذب مخمرها در غلظت-هایی که به آن حساس بودند بالاتر و در غلظتهایی که به آنها مقاوم بودند، جذب کمتری مشاهده شد. بیشترین میزان فلزات جذب شده توسط مخمر ساکارومایسز سرویزیه شامل مس (8/16 میلیگرم برگرم وزن خشک) و سرب (6/54)، در مخمر کلویورومایسز مارکسیانوس شامل مس (5/75) و سرب (4/67)، و در مخمر یاروویا لیپولیتیکا شامل مس (69) و سرب (6/69) بود. در آزمایش بررسی اثر گلوکز روی جذب در غلظتهای پایین مشاهده شد که گلوکز روی جذب سطحی بیاثر است ولی روی جذب داخلی موثر و باعث افزایش آن میگردد، زیرا جذب داخلی فرآیندی وابسته به انرژی و جذب سطحی مستقل از انرژی است.

سیستم ایمنی انسان شامل انواع متنوعی از سلول¬ها و مولکول¬ها می¬باشد که در ارتباط تنگاتنگ با سایر سیستم¬های بدن می¬باشند. مقادیر افزاینده¬ی داده¬های تولید شده در دوره¬ی پساژنومیک بررسی این سیستم را پیچیده¬تر گردانده است. بنابراین نیاز به استفاده از رویکردهای محاسباتی و کامپیوتری جهت پردازش و تفسیر داده¬ها محسوس¬تر می¬باشد. ایمونوانفورماتیک به عنوان زیرمجموعه¬ای از بیوانفورماتیک رویکردی جدید با ابزارها و پایگاه¬های اطلاعاتی مختلف می¬باشد که بررسی داده¬های ایمونولوژیک حاصل از تحقیقات آزمایشگاهی را تسهیل می¬گرداند. ایمونوانفورماتیک می¬تواند محققین را در طراحی فرضیات جدید یاری نموده و دید لازم برای انتخاب آزمایشات را فراهم آورد. با در نظر داشتن این ¬ویژگی¬ها می¬توان ایمونوانفورماتیک را حوزه¬ی جدیدی دانست که پیشرفت تحقیقات ایمنی¬شناسی را میسر می¬گرداند. در این مطالعه ما به بحث در مورد انواع مختلف ابزارها و پایگاه¬های اطلاعاتی پرداخته¬ایم که در ارتباط با حوزه¬ی ایمونوانفورماتیک می¬باشند و همچنین دیدگاه¬هایی در مورد کاربردها و افق¬های آینده¬ی این حوزه ارائه داده¬ایم.

استخراج و خالص سازی پپتید ضد میکروبی جدید از گیاه عناب (Ziziphus Jujuba)
در این مطالعه ، یک پپتید ضد باکتریایی و ضد قارچی جدید با نام Snakin-Z از میوه عناب با استفاده از ژل فیلتراسیون و کروماتوگرافی مایع با کارایی بالای فاز معکوس (RP-HPLC)، خالص سازی گردید. این پپتید از 31 آمینواسید تشکیل شده است و توالی آن عبارت است از: CARLNCVPKGTSGNTETCPCYASLHSCRKYG. وزن مولکولی این پپتید 823/3308 دالتون میباشد. هم ردیفی این پپتید با پپتیدهای ضد میکروبی موجود در داده پایگاه ضد میکروبی نشان داد که این پپتید بیشترین شباهت را به Snakin-2 دارد؛ ولی این پپتید با هیچ یک از پپتیدهای کشف شده همسانی کامل ندارد. مطالعات نشان داد که Snakin-Z دارای فعالیت ضد میکروبی علیه باکتریهای گرم منفی و گرم مثبت و همچنین قارچها است. حداقل غلظت مهاری این پپتید برای فعالیت ضد میکروبی بسیار مناسب است. این پپتید بیشترین فعالیت را بر روی قارچ Candida albicans دارد، به گونهای که حداقل غلظت مهاری آن برای این قارچ، 23/8 میکروگرم بر میلی لیتر میباشد. در بین سویههای باکتریایی، Staphylococcous aureus، مقاومت بیشتری را به عملکرد این پپتید نشان میدهد و حداقل غلظت مهاری برای این سویه، 8/28 میکروگرم بر میلی لیتر میباشد. سلولهای خونی انسان به خوبی این پپتید را تحمل میکنند و این پپتید در هیچ غلظتی از خود فعالیت همولیزی نشان نمیدهد. به دلیل فعالیت ضد میکروبی موثر این پپتید بر روی پاتوژن های مختلف، به نظر میرسد که این پپتید، نامزد مناسبی برای کاربردهای درمانی در آینده باشد.

بررسی نقش هورمون GnRH در مراحل مختلف تکوین آزمایشگاهی رویان گاو
تاثیر هورمون GnRH در تنظیم و کنترل تولید مثل در حیوان ماده و نر در مطالعات کلینیکی بخوبی نشان داده شده است ولی با توجه به نقش بسیار مهم این هورمون در سیستم تناسلی حیوان، مطالعات اندکی در مورد اثر آن روی فرآیند فولیکولوژنز ، بلوغ تخمک ، لقاح تخمک و اسپرم و تکامل رویان صورت گرفته است. هدف از این تحقیق بررسی تاثیر مقادیر مختلف این هورمون در فرآیند لقاح و تکامل رویان بوده است. در این تحقیق تعداد 1091 عدد تخمک استحصال شده به روش آسپیراسیون ، به تفکیک داخل محیط های لقاح و کشت جنین حاوی غلظت های 800 ، 1000 و 1500 نانوگرم GnRH به ازای هر میلی لیتر محیط کشت قرار گرفته و با گروه شاهد مقایسه شدند. در آزمایش اول و دوم بر اساس آنالیز میکروسکوپی از نظر ظاهری افزایش میزان تقسیم و بلاستوسیست در گروه های تیمار(خصوصا در گروه های حاوی مقادیر 800 و 1000 نانوگرم GnRH به ازای هر میلی لیتر محیط کشت)نسبت به گروه شاهد مشاهده شد ولی از نظر آماری تفاوت معنی دار بین گروه های تیمار و شاهد مشاهده نشد و همچنین افزایش قابل ملاحظه در تعداد کل سلول ها و توده سلولی خارجی در گروه های تیمار نسبت به گروه شاهد مشاهده شد(0.05>p) .

هدف: دندروزومها دسته ای از ناقل های پلیمری غیر ویروسی نسبتاً جدید هستند که باعث افزایش حلالیت داروهای نامحلول یا با حلالیت ضعیف در محیط های زیستی می شوند. کورکومین یک ترکیب ضد سرطانی با حلالیت بسیار کم در محیط های آبی است که در این پژوهش با استفاده از دندروزوم به عنوان یک ناقل، انتقال آن به سلولهای سرطانی آدنوکارسینومای معده (AGS) صورت گرفته است. مواد و روش ها: در مطالعه حاضر، کورکومین فرموله شده در دندروزوم به رده سرطانی آدنوکارسینومای معده منتقل شد و تأثیر آن بر مرگ سلولی با رنگ آمیزی به کمک رنگ پروپیدیوم آیوداید (PI) و بررسی میزان بیان ژنهای Survivin وhTERT به روش RT-PCR نیمه کمی صورت گرفت. نتایج: افزایش مرگ سلولی تحت القاء کورکومین دندروزومی در مقایسه با کورکومین آزاد در سلولهای هدف به روش فلوسایتومتری و با رنگ آمیزی PI مشاهده شد. همچنین نتایج RT-PCRنیمه کمی، کاهش نسبی بیان ژن های hTERT و Survivin که از ژن های دخیل در افزایش تکثیر و بقای سلول های سرطانی می باشند، را نشان داد. بحث: از آنجاییکه در این مطالعه کورکومین دندروزومی در مقایسه با کورکومین آزاد تأثیر بیشتری بر وقوع مرگ برنامه ریزی شده در سلولهای آدنوکارسینومای معده انسان (AGS) نشان داد. بنابراین دندروزوم ناقل مناسبی برای انتقال کورکومین نامحلول به سلول بوده و از این نانوذره می توان جهت درمان و از بین بردن سلول های سرطانی بهره برد . .

شناسایی موتاسیون در اگزون های 13 و 23 و بخشی از اینترون 12 از ژن jhdm2a در انسان و ارتباط آن با ناباروری مردان
زمینه و هدف: هیستون دمتیلاز حاوی دمین (JHDM2A) JmjC یک تنظیم کننده کلیدی تغییرات اپی ژنتیکی است که در بیضه بیان می شود، برای اسپرم زایی ضروری است و به طور اختصاصی لیزین 9 هیستون3 را در حالت مونو و دی متیله، دِمتیله می کند. JHDM2A به طور مستقیم یا غیر مستقیم روی مناطق مرکزی پروموتر پروتئین های گذرای هسته ای و پروتامین ها ، که محصول آنها برای فشرده سازی کروماتین اسپرم نیاز است، اثر می¬گذارد. روش بررسی: در این مطالعه 150 مرد نابارور که در آزمایش اسپرموگرام ثابت شده آزوسپرم و اولیگوسپرم هستند، مورد مطالعه قرار گرفتند. بر اساس مطالعات بیوانفورماتیکی و با استفاده از نرم افزار Clustal میزان مشابهت ژن jhdm2a بین انسان و موش 91% تخمین زده شد. اگزون های 13 و 23 انتخاب شده و برای شناسایی موتاسیون تکثیر شدند. محصول PCR آنها با استفاده از تکنیک SSCP مورد بررسی قرار گرفت. یافته ها : در اگزون 23 هیچ تغییری در الگوی باند ها مشاهده نشد اما در دو فرد بیمار در اگزون 13 الگوهای باندی متفاوتی در SSCP در مقایسه با افراد کنترل و سایر افراد نابارور دیده شد. نتایج تعیین توالی نشان داد که در این دو فرد بیمار یک موتاسیون از نوع جانشینی →A33838C وجود دارد.نتیجه گیری: در نتیجه با شناسایی موتاسیون های موجود در این ژن، می توان از آن به عنوان یک مارکر برای شناسایی ناباروری مردان استفاده کرد.

القاء کالوس در گیاه دارویی بابا آدم (Arctium lappa L.)
باباآدم، گیاه دارویی با اهمیتی است که حاوی ترکیبات دارویی فراوانی از جمله آرکتین و آرکتیژنین می¬باشد که خواص ضد سرطانی این متابولیت¬های ثانویه شناخته شده است. پیشرفت در روش¬های کشت بافت و باززایی این گیاه در شرایطin vitro به منظور آسان نمودن روش¬های انتقال ژن به این گیاه دارویی بسیار مهم می¬باشد. در این تحقیق، برای کالوس¬زایی در گیاه باباآدم، از ریزنمونه¬های برگ، دمبرگ و ریشه گیاهچه¬های 2 هفته¬ای استفاده شد. این ریزنمونه¬ها برای کالوس¬زایی در محیط کشت MS حاوی 5/0 درصد PVP (به عنوان آنتی¬اکسیدان)، و ترکیبی از غلظت¬های هورمونی مختلف BA و2,4-D قرار گرفتند. این آزمایشات نشان داد که از نظر تیپ ریزنمونه، ریزنمونه¬های دمبرگ بیشترین کالوس¬زایی را داشتند. از نظر غلظت¬های هورمونی، ترکیب دو هورمون در غلظت-های mg/L BA 5/0+ mg/L 2,4-D 1 و همچنینBA mg/L 1+ mg/L 2,4-D 2 بیشترین میزان تولید کالوس را نشان دادند و در نتیجه بهترین تیمارها برای القاء کالوس در گیاه دارویی باباآدم بودند.

شناسایی نشانگرهای ISSR پیوسته با صفات مورفولوژیک در جمعیتهای یونجه زراعی (Medicago sativa L.)
به منظور شناسایی نشانگرهای مرتبط با صفات ارتفاع بوته، میزان کلروفیل، وزن تر کل، وزن خشک کل، وزن تر برگ، وزن خشک برگ، وزن تر ساقه، وزن خشک ساقه، تعداد برگ، تعداد ساقه، تعداد برگ به تعداد ساقه، وزن خشک برگ به وزن خشک ساقه و وزن خشک کل به وزن تر کل در جمعیتهای مختلف یونجه زراعی، از نشانگرهای ISSR (Inter simple sequence repeat) استفاده شد. 16 آغازگر مورد استفاده، 117 مکان در 80 ژنوتیپ یونجه تولید کرد. میانگین تعداد مکانها 3/7 مکان برای هر آغازگر بود. میانگین میزان اطلاعات چند شکل (Polymorphism information content) (PIC) برای آغازگرها 77/0 و از 65/0 (UBC849) تا 93/0 (443) متغیر بود. برای شناسایی نشانگرهای مثبت مرتبط با صفات مورفولوژیک، تجزیه رگرسیون گام به گام بین دادههای ملکولی به عنوان متغیرهای مستقل و صفات مورفولوژیک به عنوان متغیرهای وابسته انجام گرفت. نشانگرهای مرتبط با صفات وزن خشک کل و وزن خشک برگ یکسان بودند. نشانگرهای مرتبط با صفات تعداد برگ و نسبت وزن خشک برگ به وزن خشک ساقه به ترتیب Kourda و UBC849 بودند. بیشترین تغییرات مربوط به صفت میزان کلروفیل (82 درصد) توسط نشانگرهای A12، UBC812، UBC808، UBC849، 425، 456 و 848 تبیین شد، در حالیکه آغازگرهای 425 و UBC808، 15 درصد از تغییرات مربوط به صفت وزن تر برگ را تبیین کردند. اکثر آغازگرهای مورد استفاده روی صفات مورد مطالعه مؤثر بودند، بنابراین میتوان از این آغازگرها همراه با اطلاعات صفات مورفولوژیک در اصلاح یونجه زراعی جهت شناسایی والدین مناسب برای تهیه جمعیتهای نقشهیابی و تولید ارقام هیبرید استفاده کرد.

شناسایی و بررسی برخی فعالیت های آنتی میکروبی باکتری بومی Enterobacter sp. DGH3
مرکبات نقش بسیار مهمی را در اقتصاد هر کشور برعهده دارند و ایران نیز از این قاعده مستثنی نیست تا جایی که باعث خودکفایی اقتصادی در زمینه کشاورزی شده است. به¬همین دلیل لازم است که از بروز آفات تا حد امکان جلوگیری بعمل آید و در جهت کنترل آن ها اقدامات اساسی انجام داد. بیماری های باکتریایی و قارچی گوناگونی وجود دارند که اثرات مخربی در صنعت کشاورزی و بخصوص مرکبات وارد می نمایند. کنترل بیماری های باکتریایی،تنها از طریق سموم و آنتی بیوتیک ها و آن هم بطور مقطعی امکان پذیر می باشد. استفاده از ترکیبات آنتی باکتریال جهت کنترل این این بیماری ها می تواند بسیار مفید واقع شود. باکتری Enterobacter sp. DGH3، نوعی باکتری بومی می باشد که قادر است باکتری عامل بیماری شانکر در مرکبات را از بین ببرد. امید است که با بهینه سازی محیط کشت این باکتری و همچنین بهینه سازی تولید ترکیب آنتی باکتریال توسط باکتری مذکور بتوان گام مهمی در جهت کنترل این بیماری برداشت.

ارتباط پلی مورفیسم G/T 317- پروموتر ژن Muc6 با افزایش خطر ابتلا به بیماری زخم پپتیک
چکیدهموسین ها (Muc) گلیکوپروتئین هایی با وزن مولکولی بالا هستند که بخش اصلی لایه ژله ای موکوس را تشکیل داده و در حفاظت از اپی تلیوم معده ای-روده ای نقش دارند. با توجه به بیان زیاد و اختصاصی Muc6 در معده و دوازدهه، تغییر بیان Muc6 در بیماری زخم پپتیک و نقش پلی مورفیسم های پروموتر در بیان ژن ها، در این مطالعه ارتباط پلی مورفیسم پروموتر ژن Muc6 با خطر ابتلا به بیماری زخم پپتیک با تکنیک PCR و توالی یابی بررسی شد. نتایج نشان داد که فراوانی آلل T در جایگاه 317- پروموتر ژن Muc6 در افراد بیمار در مقایسه با افراد کنترل به طور معنی داری بیشتر است (005/0>P ,1=df ,94/8=2χ) به طوریکه اگر آلل T به صورت غالب در نظر گرفته شود ژنوتیپ های TG+TT در مقایسه با GG، خطر ابتلا به بیماری زخم پپتیک را بیش از سه برابر افزایش می دهد (01/0>P ,6/8-28/1=95% CI ,3/3=OR).

فلاو آنزیم پرولین دهیدروژناز (EC 1.5.99.8) نقش مهمی را در زیست حسگرها و کیتهای غربالگری به منظور تشخیص بیماریهای متابولیک مرتبط با نقص پرولین ایفاء می نماید. هدف از این تحقیق، جداسازی و تعیین مشخصات میکروارگانیسم های تولید کننده آنزیمهای دخیل در متابولیسم پرولین از نمونه های خاک ایران بود. غربالگری آنزیم های تخریب کننده L-پرولین از نمونه های خاک در سه مرحله شامل تکنیک کشت غنی، کروماتوگرافی لایه نازک و سنجش فعالیت آنزیمی، انجام شد. میکروارگانیسم جداسازی شده توسط واکنشهای بیوشیمیایی، آنالیز ژنهای RNA ریبوزومی s16، 16S-23S rRNA Internal Transcribed Spacer (ITS) و رسم درخت فیلوژنتیکی تعیین مشخصات گردید. مطالعات کینتیکی بر روی آنزیم شناسایی شده انجام شد و ساختار سه بعدی آن توسط روشهای بیوانفورماتیکی، شبیه سازی گردید. در بین 150 میکروارگانیسم جداسازی شده از 30 نمونه خاک، تنها یک سویه باکتری، فعالیت قابل ملاحظه ای نسبت به اسید آمینه L-پرولین (U/ml10) از خود نشان داد. پارامترهای ثابت میکائیلیس-منتن و سرعت ماگزیمم به ترتیب 12/0 میلی مولار و 11/0 میلی مولار بر دقیقه محاسبه شدند. سویه باکتری با استفاده از آنالیز فیلوژنتیکی ژنهای RNA ریبوزومی s16 و ITS به عنوان Biotype جدیدی از سودوموناس پوتیدا شناسایی شد. ساختاز سه بعدی شبیه سازی شده از آنزیم مذکور نیز قرار داشتن پرولین دهیدروژناز جداسازی شده در آنزیمهای خانواده سودوموناس پوتیداها را تایید نمود. این تحقیق اولین گزارش از تولید پرولین دهیدروژناز توسط باکتری سودوموناس پوتیدا جداسازی شده از نمونه خاک می باشد.

مطالعه شبیه سازی دینامیک ملکولی و حرکات عملکردی آنزیم لیپازA از گونه باسیلوس سوبتیلیس
آنزیم لیپاز باسیلوس سوبتیلیس یکی از کوچک ترین پروتئین های متعلق به خانواده β/α هیدرولازها می-باشد. این آنزیم بر خلاف همه لیپازها خاصیت فعال سازی سطحی را از خود نشان نمی¬دهد ولی همانند دیگر لیپازها کاربردهای فرآوانی در زمینه¬های صنعتی، دارویی، صنایع غذایی و زیست محیطی دارا می¬باشد. تعدادی گزارش از تولید این لیپاز مهندسی شده ارائه شده که اساس طراحی در آن¬ بصورت جهش¬های تصادفی می¬باشد. لذا مطالعه دینامیک ملکولی این آنزیم در دماهای مختلف برای بررسی خصوصیات دینامیکی آن ضروری به نظر می¬رسد. شناسایی این حرکات ملکولی برای تغییر در روش طراحی از تصادفی به طراحی منطقی آنزیم بسیار موثر می¬باشند. بر اساس مطالعه شبیه سازی دینامیک ملکولی این آنزیم در مدت 15 نانوثانیه، حرکات ملکولی در لوپ¬های دوطرف جایگاه فعال مشاهده گردید که میزان آن در دماهای مختلف با هم متفاوت بودند. از سوی دیگر دو لوپ کلیدی که در ایجاد حرکات عملکردی نقش دارند در طرف دیگر آنزیم شناسایی شدند که دارای نقش لولا در محور حرکتی اجزای ساختاری دوم در اطراف جایگاه فعال می-باشند.

کلونینگ ژن آلفا آمیلاز در وکتور انتقالی pGFK114.1 اشرشیاکولی- باکتروئیدی و انجام کانژوگاسیون بین سویه های مختلفE.coli جهت امکان استفاده از آن در محیط گوارشی نشخوارکنندگان
به منظور افزایش کارآیی تولید در صنعت دامپرروری می توان از تکنولوژی DNA نوترکیب بهره بردکه پیشرفت های اخیر در سیستم انتقال ژن، تغییرات ژنتیکی باکتری های شکمبه ای را با توانایی تولید آنزیم های هیدرولیز کننده ممکن ساخته است. این تکنولوژی می تواند ضمن کاهش معایب افزودنی های آنزیمی افزایش بازدهی مواد مغذی در شکمبه را به دنبال داشته باشد. از آنجاییکه فرایند ترانسفورماسیون باکتری های سیستم گوارشی دارای بازدهی پایینی می باشد کانژوگاسیون می تواند یکی از روش های موثر انتقال ژن در شکمبه باشد. در بیشتر تحقیقات صورت گرفته از باکتری اشرشیا کولی بهره می گیرند که یک باکتری بی هوازی اختیاری است و می تواند در شرایط بی هوازی شکمبه رشد نماید. در این تحقیق ژن آلفا آمیلاز که پیش از این در وکتور pET28a کلون شده بود با تکثیر در واکنش زنجیره ای پلیمراز در وکتور انتقالی pGFK114.1 ساب کلون شد و درون سویه ای از باکتری DH5α E.coli ترانسفورم گردید. سپس با الگو گرفتن از محیط سیستم گوارش جهت انتقال ژن، عمل کانژوگاسیون بین سویه دیگری از باکتری اشرشیاکولی با کمک وکتور انتقالی pRK231 که در سویه TG1 E.coli ترانسفورم شده بود، صورت گرفت. به این ترتیب باکتری اشرشیا کولی حاوی وکتور انتقال یافته از طریق کانژوگاسیون بیانگر انتقال موفق مواد ژنتیکی توسط این روش می باشد که در محیط شکمبه هم طبق گزارشات موجود به راحتی قابل انجام می باشد.
