In past years, various physical and chemical methods were evaluated and tested to remove heavy metals, but most of them happen to be tough or hazardous, complex, incomplete, expensive moreover requiring lot of energy. Using microorganisms as adsorbents for removal of heavy metal offers a potential alternative to mentioned methods. In this study, three standard yeast including Saccharomyces cerevisiae PTCC 5052, Kluyveromyces marxianus DSMZ5422 Yarrowia lipolytica DSMZ 8218, was used. Resistance of yeast to different concentrations of copper and lead were measured, The yeast were more resistant towards copper (up to 6mM by Y. lipolytica, 5mM by K. marxianus and 1mM by S. cerevisiae) and less resistant towards lead (up to 2mM by the used yeasts). Then based on the observed resistance of the yeast, their ability in metal absorption was examined. Yeast metal uptake at concentrations that they were sensitive, were more. The maximum uptake by the yeasts were 168mg/ g dry weight (g dry W) copper and 54.6mg/g dry W lead by S. cerevisiae, 75.56mg/g dry W from copper and 67.46mg/g dry W from lead by K. marxianus, and 696mg/g dry W copper and 69.66mg/g dry W from lead by Y. lipolytica. Another test was performed to investigate the effect of glucose on uptake in low concentrations. It was observed that glucose was ineffective on absorption which is energy-independent and effective on bioaccumulation which is energy-dependant.}, keywords = {Key words: Yeast,Heavy metals,Metal resistance,Biosorption,Atomic absorption}, title_fa = {بررسی تحمل و جذب فلزات مس و سرب توسط سه سویه استاندارد مخمر}, abstract_fa = {آلودگی محیط زیست توسط فلزات سنگین به یکی از جدی‌ترین معضلات زیست محیطی امروز تبدیل شده است. در طی سالیان گذشته روش‌های مختلف فیزیکی و شیمیایی برای پاکسازی فلزات سنگین مورد بررسی قرار گرفته‌اند، اما اکثر آنها مشکل، گران و انرژی بر هستند. استفاده از میکروارگانیزم‌ها برای جذب فلزات سنگین، می‌تواند جایگزینی مناسب برای این روش‌ها باشد. در این تحقیق از سه مخمر استاندارد PTCC 5052 Saccharomyces cerevisiae، Kluyveromyces marxianus DSMZ 5422،Yarrowia lipolytica DSMZ 8218، برای بررسی مقاومت نسبت به غلظت‌های مختلف فلزات سنگین مس و سرب استفاده شد. مقاومت بیشتری نسبت به مس (یاروویا لیپولیتیکا تا 6 میلی‌مول، کلویورومایسز مارکسیانوس تا 5 میلی‌مول و ساکارومایسز سرویزیه تا 1 میلی‌مول) و مقاومت کمتری نسبت به سرب (هر سه مخمر تا غلظت 2 میلی‌مول) مشاهده شد. سپس بر اساس مقاومت‌های مشاهده شده توانایی مخمرها در جذب این فلزات سنگین مورد آزمایش قرار گرفت. میزان جذب مخمرها در غلظت-هایی که به آن حساس بودند بالاتر و در غلظت‌هایی که به آن‌ها مقاوم بودند، جذب کمتری مشاهده شد. بیشترین میزان فلزات جذب شده توسط مخمر ساکارومایسز سرویزیه شامل مس (8/16 میلی‌گرم برگرم وزن خشک) و سرب (6/54)، در مخمر کلویورومایسز مارکسیانوس شامل مس (5/75) و سرب (4/67)، و در مخمر یاروویا لیپولیتیکا شامل مس (69) و سرب (6/69) بود. در آزمایش بررسی اثر گلوکز روی جذب در غلظت‌های پایین مشاهده شد که گلوکز روی جذب سطحی بی‌اثر است ولی روی جذب داخلی موثر و باعث افزایش آن می‌گردد، زیرا جذب داخلی فرآیندی وابسته به انرژی و جذب سطحی مستقل از انرژی است.}, keywords_fa = {کلمات کلیدی: مخمر,فلزات سنگین,جذب زیستی,مقاومت به فلزات سنگین,جذب اتمی}, url = {https://cell.ijbio.ir/article_320.html}, eprint = {https://cell.ijbio.ir/article_320_6ca60996192eac272f4184e0321debb6.pdf} }
