In this research, 71 agricultural soil samples collected from suburb of Ray and Karaj cities. Screening of bacteria on semiselective SX agar medium, performing morphological and biochemical tests and evaluation of two specific virulence factors resulted in a number of X. campestris isolates among all putative growing bacteria. Relative percentage of X. campestris to total number of bacteria grown on SX agar was 0.69%. Capability of exopolysaccharide production by average concentration of 10.98 g l-1 and average viscosity of 1403 cP and presence of xanthomonadin pigment with maximum absorbance index within the range of 441.0-444.7 nm wavelengths, as two main virulence factors, was determined and confirmed. Molecular identification for 25% of the randomly selected isolates possessing virulence factors, indicated X. campestris species by confirming the presence of specific hrcC gene marker. This study indicates that despite lack of any report on prevalence of the related diseases, the potentially virulent X. campestris can be found in soil of cabbage farms in suburb of Ray and Karaj cities. As a case study, existence of these strains in soil is noticeable because of probability of losses caused by spoilage of agricultural products which is prevalent in Iran.}, keywords = {cabbage (Brassica oleracea),soil,Xanthomonas campestris,exopolysaccharide,xanthomonadin}, title_fa = {باکتریهای بالقوه بیماریزای Xanthomonas campestris در خاک کشتزارهای ری و کرج}, abstract_fa = {باکتریهای جنس Xanthomonas از مهم ترین باکتریهای بیماریزای گیاهان زراعی بوده و از عوامل مهم فساد پس از برداشت و خسارت اقتصادی می‌باشند. در این مطالعه 71 نمونه از خاک کشتزارهای حومة شهرهای ری و کرج جمع‌آوری شد. غربالگری باکتریها بر روی محیط نیمه‌انتخابی SX agar، انجام آزمونهای ریخت‌شناسی و بیوشیمیایی و بررسی دو شاخص ویژة ویرولانس منجر ‌به جداسازی تعدادی جدایة X. campestris در میان باکتریهای رشد یافته احتمالی شد. تراکم نسبی X. campestris در مقایسه با تراکم کل باکتریهای رشد یافته بر‌روی محیط SX agar، 69/0 درصد بود. قابلیت تولید اگزوپلی‌ساکارید با میانگین تراکم g l-1 98/10 و میانگین ویسکوزیتة cP 1403 و نیز وجود پیگمان زانتومونادین با شاخص جذب نوری بیشینه در طول موجهای nm 7/444- 0/441 به عنوان دو شاخص اصلی‌ ویرولانس این باکتریها مورد بررسی و تأیید قرار‌گرفت. شناسایی مولکولی 25 درصد از جدایه‌های واجد فاکتورهای ویرولانس که بطور تصادفی انتخاب شدند، تعلق به گونة X. campestris را به واسطة وجود مارکر ژنی اختصاصی hrcC تأیید کرد. این بررسی نشان می‌دهد که علی‌رغم نبود گزارشی از شیوع بیماری، باکتریهای X. campestris بالقوه بیماریزا در خاک کشتزارهای کلم در حومة ری و کرج یافت می‌شوند. به عنوان یک بررسی موردی، وجود این سویه‌ها در خاک به دلیل ایجاد خسارتهای ناشی از فساد محصولات که در ایران شایع است، حائز اهمیت می‌باشد. }, keywords_fa = {کلم (Brassica oleracea),خاک,Xanthomonas campestris,اگزوپلی‌ساکارید,زانتومونادین}, url = {https://cell.ijbio.ir/article_207.html}, eprint = {https://cell.ijbio.ir/article_207_e790bd2718d77f040f8c2ead0f1c0edb.pdf} }
