According to studies, miRNA-34a (miR-34a) expression declines in colon cancer. The aim of this study was to investigate the effect of miR-34a on growth and migration inhibition and apoptosis induction of colon cancer cells.In this experimental study, HCT116 human colorectal cancer cells were used in RPMI1640 medium. MiR-34a transmission into cancer cells was performed by the jetPEI transfection reagent. MTT assay to determine the survival of cancer cells and the qRT-PCR test to confirm the transfer of miR-34a to the cells and also an investigation of the expression of Caspase genes were used. Finally, the Wound healing test in order to investigate the migration status of transfected and control cells was performed. The expression changes of the intended genes in the transfected cells were compared with the control cells using the 2-ΔΔCt method.The MTT test showed that induction of miR-34a causes the death of colorectal cancer cells. Also, the results of the qRT-PCR test revealed a significant increase of miR-34a in the HCT116 transfected cells compared to the control as well as increased expression of Caspase genes in transfected cells. Ultimately, the results of the Wound healing test showed reduced migration of transfected cells compared to the control cells. The study showed that miRNA-34a plays an important role in inhibiting the growth and migration of colon cancer cells and can be a candidate for molecular therapies.}, keywords = {miRNA-34a,Colon cancer,HCT 116,cell migration,caspase}, title_fa = {اثر جایگزینی miRNA-34a در مهار رشد و مهاجرت سلولی و افزایش بیان ژن های کاسپازی در رده سلولی HCT116 سرطان کلون}, abstract_fa = {سرطان کلون به عنوان شایع ترین سرطان دستگاه گوارش، چهارمین علت مرگ و میر در جهان است. بر اساس مطالعات انجام یافته، بیان miRNA-34a (miR-34a) در سرطان کولون کاهش می یابد. هدف این مطالعه، بررسی اثر جایگزینی miR-34a در مهار رشد و مهاجرت سلولی و القای آپوپتوز سلول های سرطان کلون می باشد. در این مطالعه تجربی از سلول‌های سرطانی HCT116 در محیط کشت RPMI1640 استفاده شد. انتقال miR-34a به داخل سلول‌های سرطانی توسط ریجنت jetPEI صورت گرفت. تست MTT برای بررسی میزان زنده بودن سلول‌های سرطانی و تست qRT-PCR برای تایید انتقال miR-34a به داخل سلول ها و نیز بررسی بیان ژن های کاسپازی استفاده شد. در نهایت، تست Wound healing به‌منظور بررسی وضعیت مهاجرت سلول های ترانسفکت شده و کنترل انجام یافت. میزان تغییرات بیان ژن های مورد نظر در سلول های ترانسفکت شده نسبت به سلول های کنترل با روش 2-∆∆Ct بررسی شد. تست MTT نشان داد که القاء miR-34a به‌صورت وابسته به دوز باعث مرگ سلول‌های سرطان کلورکتال می شود. همچنین نتایج حاصل از تست qRT-PCR افزایش قابل‌توجه miR-34a را در سلول‌های HCT116 ترانسفکت شده نسبت به کنترل و همچنین افزایش بیان ژن های کاسپاز 3، 8 و 9 را در سلول های ترانسفکت شده نشان داد. در نهایت نتایج تست Wound healing کاهش مهاجرت سلول‌های ترانسفکت شده را در مقایسه با سلول‌های کنترل نشان داد. این مطالعه نشان داد که miRNA-34a نقش مهمی در مهار رشد و مهاجرت سلول های سرطان کولون دارد و می تواند کاندیدای مناسبی برای درمان های مولکولی باشد.}, keywords_fa = {miRNA-34a,سرطان کولون,HCT 116,مهاجرت سلولی,کاسپاز}, url = {https://cell.ijbio.ir/article_1632.html}, eprint = {https://cell.ijbio.ir/article_1632_40ac48775b75e1381e99ea90ce2aaf9f.pdf} }
