Application of the genes encoding glyphosate degrading enzymes such as glyphosate oxidoreductase (GOX) in combination with a glyphosate-tolerant EPSPS is the ultimate approach to provide commercial rates of glyphosate tolerance. The gox gene was first isolated from Ochrobactrum antrophi strain LBAA that catalyses the cleavage of glyphosate into aminomethylphosphonic acid and glyoxalate. In this study, the gox gene was synthesized and cloned in pET28a expression vector and transformed in E. coli. The expression of the target protein was confirmed by SDS-PAGE. The experiment for optimization of gene expression showed that the optimal condition was 37oC for bacterial culture, 4 hours of induction time using 1 mM IPTG in OD600=0.6. In the next step, in order to perform a bioassay on transformed bacteria, the threshold of tolerance of control bacteria in minimum phosphate-free medium with different concentrations of glyphosate was investigated and compared with recombinant bacteria. The result indicated that the wild type bacteria were not able to tolerate concentration of more than 0.5 mM glyphosate but the recombinant bacteria survived to a concentration of 1 mM. These results were confirmed by colony counting on selective media with three repetitions. Hence, glyphosate oxidoreductase gene could be a suitable candidate besides the other glyphosate resistance genes for genetic manipulation of strategic plants with the aim of obtaining higher and more sustainable levels of resistance to this herbicide.}, keywords = {Glyphosate,Gene expression,synthetic Glyphosate oxidoreductase gene}, title_fa = {افزایش تحمل به علف کش گلایفوسیت در نتیجه بیان ژن نوترکیب گلایفوسیت اکسیدورداکتاز (gox) در باکتری اشرشیا کولی}, abstract_fa = {متحمل‌سازی گیاهان زراعی نسبت به علف‌کش گلایفوسیت از طریق دست‌ورزی‌های ژنتیکی یکی از کاراترین روش‌های موجود در مدیریت علف‌های هرز محسوب می‌شود. راه حل مطلوب برای توسعه گیاهان متحمل در سطح تجاری، به کارگیری ژن‌های عامل آنزیم های تجزیه کننده گلایفوسیت نظیر گلایفوسیت اکسیدورداکتاز در کنار EPSPS مقاوم به گلیفوسیت می‌باشد. ژن گلایفوسیت اکسیدورداکتاز اولین بار از باکتری Ochrobactrum antrophi سویه LBAA جدا گردیده که آنزیم آن، علف‌کش گلایفوسیت را به آمینومتیل فسفونیک اسید و گلی‌اکسالات تبدیل می‌کند. در تحقیق حاضر، برای مطالعه کارایی گلایفوسیت اکسیدورداکتاز، توالی ژن آن سنتز و در ناقل بیانی pET28a همسانه سازی گردید. پس از انتقال به باکتری اشرشیاکلی، بیان پروتئین هدف با روش SDS-PAGE مورد تائید قرار گرفت. آزمایشات بهینه سازی بیان ژن در باکتری نشان داد که شرایط بهینه شامل دمای 37 درجه سانتیگراد برای کشت، 4 ساعت پس از القا با IPTG یک میلی‌مولار در OD600=0.6 می‌باشد. در مرحله بعد، به منظور انجام آزمون زیستی باکتری‌های تراریخت، حد آستانه تحمل باکتری‌های شاهد در محیط حداقل فاقد فسفات با غلظت‌های مختلف گلایفوسیت انجام و با باکتری‌های نوترکیب مقایسه گردید. نتایج نشان داد که باکتری‌های شاهد غلظت بیشتر از 5/0 میلی‌مولار گلایفوسیت را نمی‌توانند تحمل کنند؛ در صورتی که باکتری‌های نوترکیب تا غلظت 1 میلی‌مولار زنده می‌مانند. این نتایج با انجام روش شمارش کلونی و پس از سه تکرار تایید گردید. بنابراین ژن گلایفوسیت اکسیدورداکتاز می‌تواند کاندید مناسبی در کنار سایر ژن‌های عامل مقاومت به گلایفوسیت جهت دست‌ورزی ژنتیکی گیاهان استراتژیک با هدف بدست آوردن سطوح بالاتر و پایدارتر مقاومت به این علف‌کش باشد.}, keywords_fa = {گلایفوسیت,بیان ژن,ژن مصنوعی گلایفوسیت اکسیدورداکتاز}, url = {https://cell.ijbio.ir/article_1585.html}, eprint = {https://cell.ijbio.ir/article_1585_05c802aff5dba1df839a215d04d6604c.pdf} }
