The aim of this study was to produce phytase from rice bran extract as a rich and inexpensive source of phytic acid using Bacillus isolated from sediments of the Caspian Sea. Using thermal treatment of the sediment samples, phytase producing Bacillus isolates were screened on PSM agar. In addition, after a quantitative assay of phytase production using the ammonium molybdate method, phytase production of the isolates was investigated in the MGE broth containing rice bran extract. Screening results of 40 isolates demonstrated that the four isolates MGH2, MGH3, MGH4 and MGH7 showed high levels of phytase production with a clear halo zone forming around the colony. Morphological and physiological characteristics indicated that all of the selected isolates belong to Bacillus. Maintaining a clear halo zone after staining with ammonium molybdate confirmed the production of phytase in all selected isolates. In addition, the results of the quantitative assay of phytase production in the isolates showed that maximum and minimum phytase production relate to MGH4 with 10.58 U ml-1 and MGH3 with 9.34 U ml-1, respectively. The results showed an increase of 55-60 percent of phytase production in MGE broth compared to PSM broth. The results of present study indicated that rice bran extract can be considered as a suitable and affordable substrate with high levels of phytase production.}, keywords = {Phytase,rice bran extract,Bacillus,Caspian Sea}, title_fa = {تولید فیتاز از عصاره سبوس برنج به کمک سویه‌های باسیلوس جداشده از رسوبات بستر دریای مازندران}, abstract_fa = {فیتیک اسید فرم اصلی ذخیره‌ای فسفر است که برای حیوانات تک معده نظیر ماکیان و ماهی‌ و حتی انسان قابل هضم نیست. هدف از این تحقیق بررسی تولید فیتاز از عصاره سبوس برنج به عنوان منبع غنی و ارزان قیمت فیتات توسط سویه‌های باسیلوس جداشده از رسوبات بستر دریای مازندران بود. با تیمار حرارتی نمونه‌های رسوب بستر دریا، جدایه‌های باسیلوس تولید کننده فیتاز توسط محیط‌ کشت PSM آگار غربالگری شدند. همچنین پس از سنجش کمی تولید آنزیم فیتاز به روش آمونیوم مولیبدات، تولید فیتاز جدایه‌ها در محیط کشت مایع MGE حاوی عصاره سبوس برنج بررسی شد. نتایج غربالگری 40 جدایه نشان داد که چهار جدایه MGH2، MGH3، MGH4 و MGH7 با هاله شفاف در اطراف کلنی، بیشترین توانایی تولید فیتاز را داشتند. بررسی مشخصات مورفولوژیکی و فیزیولوژیکی نشان داد که تمام جدایه‌های منتخب متعلق به جنس باسیلوس بودند. حفظ هاله شفاف پس از رنگ آمیزی با آمونیوم مولیبدات، نشان داد که تمام جدایه‌های منتخب توانایی تولید فیتاز را داشتند. همچنین نتایج سنجش کمی تولید فیتاز توسط جدایه‌ها نشان داد که بیشترین و کمترین مقدار تولید فیتاز به ترتیب مربوط به جدایه MGH4 با U ml-110.58 و جدایه MGH3 با U ml-19.34 بود. نتایج تولید فیتاز با استفاده از عصاره سبوس برنج در محیط رشد مایع MGE، افزایش 60-55 درصدی نسبت به محیط رشد مایع PSM را نشان داد. نتایج پژوهش حاضر نشان داد که عصاره سبوس برنج می‌تواند به عنوان سوبسترای مناسب و مقرون به صرفه با سطح تولید فیتاز بالا در نظر گرفته شود.}, keywords_fa = {فیتاز,عصاره سبوس برنج,باسیلوس,دریای مازندران}, url = {https://cell.ijbio.ir/article_1277.html}, eprint = {https://cell.ijbio.ir/article_1277_0473147b025df569350cccb809046d0a.pdf} }
