Interaction effect of optical density of Agrobacterium, Agrobacterium strain and type of Agrobacterium killing antibiotic on number of regenerated plant was significant at 1% probability level and less inhibitory effect on regenerated plant was related to OD600=0.6-0.8×GV3101 Agrobacterium starin×160 mg/L of timentin antibiotic. More regenerated plant resistance to hygromaycin was related to 254.8 µm/L of acetosyringone. More regenerated plant was achieved from 30 minutes inoculation duration than 1and 20 minutes inoculation duration, nonetheless there was no statistically significant difference between means of regenerated plant from 20 and 30 minutes inoculation duration. The presence of the target gene was evaluated using PCR analysis on regenerated plants. Results of PCR analysis confirm entity of at least one copy of the target gene on plants genome. The superiority of transgenic plants against non-transformed plants for tolerance to -0.3 Mpa osmotic stress was proved by osmotic stress tolerance bioassay.}, keywords = {Agrobactrium,Ajowan,Flavodoxin,Hygromaycin,Transformation Factors}, title_fa = {بهینه کردن انتقال ژن به واسطه اگروباکتریوم در گیاه دارویی زنیان با استفاده از ژن fld}, abstract_fa = {به منظور بهینه کردن شرایط انتقال ژن به گیاه دارویی زنیان عوامل مختلف شامل رقت آگروباکتریوم، سویه آگروباکتریوم، نوع آنتی بیوتیک کشنده آگروباکتریوم، غلظت استوسرینگون و مدت زمان تلقیح بر روی تعداد گیاهان باززا شده با استفاده ازسازه ژنی p6U-Ubi-FVT دارای ژن انتخابیHPT و ژن fld بررسی شد. غلظت 20 میلی گرم در لیتر هیگرومایسین برای غربالگری سلول های تراریخت و جلوگیری از تولید کالوس در سلول های غیر تراریخت بهترین غلظت تشخیص داده شد. اثر متقابل سه عامل تراکم سلولی آگروباکتریوم، سویه آگروباکتریوم و نوع آنتی بیوتیک کشنده آگروباکتریوم بر تعداد گیاهان باززا شده در سطح احتمال یک درصد معنی دار بود و رقت 6/0-8/0آگروباکتریوم × سویه GV3101 آگروباکتریوم × غلظت 160 میلی گرم در لیتر آنتی بیوتیک کشنده تیمنتین اثر بازدارنده کمتری بر تعداد گیاهان باززا شده داشتند. بیشترین تعداد گیاهان باززا شده مقاوم به آنتی بیوتیک هیگرومایسین مربوط به غلظت 8/254 میکرومولار در لیتر استوسرینگون بود. گیاهان باززا شده بیشتری در مدت زمان تلقیح 30 دقیقه نسبت به زمان های تلقیح 20 دقیقه و 1 دقیقه حاصل شد همچنین تفاوت آماری معنی داری بین میانگین تعداد گیاهان باززا شده حاصل از 20 دقیقه و 30 دقیقه زمان تلقیح مشاهده نشد. بررسی حضور ژن مورد نظر با استفاده از واکنش زنجیره ای پلی مراز روی گیاهان باززا شده انجام شد. نتایج وجود حداقل یک نسخه از ژن هدف را در ژنوم گیاهان تائید کرد. برتری گیاهان تراریخت نسبت به گیاهان غیرتراریخت در تحمل به تنش اسمزی 3/0- مگاپاسکال توسط آزمون زیست سنجی تحمل به تنش اسمزی تایید گردید.}, keywords_fa = {آگروباکتریوم,زنیان,فاکتور های تراریختی,فلاودوکسین,هیگرومایسین}, url = {https://cell.ijbio.ir/article_1102.html}, eprint = {https://cell.ijbio.ir/article_1102_4a028d61670f4defdccb294e6e865ec1.pdf} }
