زرین گیاه با نام علمی (Dracocephalum kotschyi Boiss) گیاهی علفی، چندساله و اندمیک ایران از خانواده نعناع است. تحقیقات اخیر دارویی نشان داده است که فلاونوئید متوکسی موجود در قسمتهای مختلف گیاه خاصیت ضدسرطانی دارد. ریشههای مویین پس از تلقیح ریزنمونه برگی با سویه 15834 باکتری آگروباکتریوم رایزوژنز بدست آمد. در این مطالعه اثر محرکهای زیستی شامل محرک کلشیسین در غلظتهای (0، 01/0، 03/0 و 05/0 درصد) و کیتوسان در غلظتهای (0، 50، 100 و 150 میلیگرم در لیتر) به مدت 48 ساعت بر رشد ریشههای مویین تولید شده و تولید رزمارینیک اسید مطالعه شد. آنالیز واریانس دادههای حاصل از اثر کلشیسین و کیتوسان بر میزان فعالیت آنتی اکسیدانی کل، فنل کل و میزان فلاونوئید نشان داد که بین تیمارها اختلاف معنیدار در سطح (P 0.01) قابل مشاهده میباشد. بیشترین افزایش در میزان آنتی اکسیدان مربوط به سطح 05/0 درصد میباشد (33/90 درصد) و کم ترین میزان فعالیت آنتی اکسیدانی نیز در ریشههای غیر تراریخت (63 درصد) مشاهده گردید. بیشترین افزایش در میزان آنتی اکسیدان مربوط به سطح 05/0 درصد میباشد (33/90 درصد). نتایج آنالیز HPLC نشان داد که بیشترین مقدار رزمارینیک اسید در ریشههای تراریخت تیمار شده با محرک کلشیسین با غلظت 05/0 درصد به میزان µg /mg DW 8/35 و کیتوسان با غلظت mg/l 100، به میزان µg /mg DW 1/15 و کمترین مقدار رزمارینیک اسید مربوط به ریشههای غیر تراریخت (شاهد)، µg /mg DW 8/13 و ثبت گردید.
Dracocephalum kotschyi Boiss belonging to Labiateae family is an herbaceous, pereninal and endemic plant known in Iran as Zarrin-Giah. Recent pharmacological studies demonstrated this plants with methoxylated flavonoids, have anti-cancer effects. Hairy roots were obtained from leaves explants, inoculated with 15834 strain of Agrobacterium rhizogenesis. In present study, The effects of biotic elicitors including, Colchicine with different cocentrations ( 0, 0.01, 0.03 and 0.05% (w/v)) and Chitosan (0, 50, 100 and 150 mg/l) for 48 hours on hairy root growth and Rosmarinic acid production were investigated. ANOVA results of Antioxidant activity, total phenol and total flavonoids showed significant difference among the various treatments (P<0.01). maximum Antioxidant activity (90.23%) was observed in 0.05% colchicine and lowest Antioxidant activity (63%) was observed in non-transformed roots. HPLC analysis revealed that the maximum Rosmarinic acid content (35.8 and 15.1 µg /mg DW ) obtained in colchicine (0.05%) and chitosan (100 mg/l) respectively. Although minimum Rosmarinic acid content (13.8 µg /mg DW) were recorded in non-transformed roots. based on the importance of rosmarinic acid production in the pharmaceutical industry, the results showed that using biotechnological methods, production improving of valuable medicinal substances is acceptable.
